ABOUT DOCTORAL WORK
During Ph.D. tenure, my specific field of research work was aimed to infer genetic
diversity among three species namely Ompok pabda, O. pabo and O. bimaculatus using three
mitochondrial (COI, Cyt b and 16S rRNA) genes.
Fishes are probably more diverse at all taxonomic levels and have more species than all
other vertebrate group combined together in patterns of abundance, diversity and distribution. Catfishes
are important part of the fish fauna that exists in wetlands and many of them are economically important
with high nutritive value. There are about 158 species of inland catfishes from 50 genera and 13 families
in India. The catfish genus Ompok (Lacepede, 1803) refers to medium-sized Silurids that are usually
found in lakes and large rivers throughout South and Southeast Asia. Four species belonging to genus
Ompok (O. pabda, O. pabo, O. bimaculatus and O. malabaricus) have been reported from India. Among
them, O. pabda, O. pabo and O. bimaculatus have been categorized as endangered fish species in India.
Identification and characterization of fish species are usually based on morphological
characters that are sometimes found to be erroneous. Synonymous citation in FishBase indicates the
possibility of ambiguous taxonomic identifications of Ompok species. Hence, it is required to examine
systematic relationships among the species using molecular marker to provide supplementary
information for taxonomic identification.
Mitochondrial DNA markers cytochrome c oxidase I (COI), cytochrome b (Cyt b) and 16S
ribosomal RNA (16S rRNA) genes are excellent markers for examining biogeographical events and
phylogenetics. COI gene has been studied for discriminating between closely related species across
diverse animal phyla and this has been used for marine and freshwater fishes. Cyt b gene has been
widely used to study genetic variation, phylogenetic relationships and taxonomy in many fishes and
invertebrates. The mtDNA 16S rRNA gene has been used for establishing the evolutionary relationship
of lineages of similar divergence in many fishes. Hence, studies using three mitochondrial genes (COI,
Cyt b and 16S rRNA) was taken up to resolve the taxonomic ambiguities.
A total of one hundred and six sequences of each mtDNA genes were generated from O.
pabda, O. pabo and O. bimaculatus. Twenty one haplotypes of COI gene, twenty haplotypes of Cyt b
gene and thirty haplotypes of 16S rRNA gene were observed. The sequences were bidirectionally
aligned and submitted to NCBI for GenBank Accession Number (COI-FJ229970 - FJ230075; Cyt b-
FJ711226 - FJ711331 and 16S rRNA- GQ469537 - GQ469642). Simplicity and un-ambiguity were
observed among all the sequences. The sequences were analyzed for species identification &
phylogenetic inference.

Neighbor-Joining (NJ) phylogenetic tree was constructed using K2P distance and
Maximum Parsimony (MP) tree was constructed using Close-Neighbor-Interchange algorithm. NJ and
MP phylogeny trees constructed based on three mitochondrial gene (COI, Cyt b and 16S rRNA)
sequences of three Ompok species showed identical phylogenetic trees. The Ompok species namely O.
pabda and O. bimaculatus which were morphologically & meristically more closer, formed one cluster
whereas O. pabo formed a separate cluster. In conclusion, the results were congruent with the taxonomic
identification of Ompok species based on morphological characters.
The present study has strongly supported the utility of mitochondrial markers for
estimating genetic diversity and establishing phylogenetics relationship among three Ompok species (O.
pabda, O. pabo and O. bimaculatus). The partial sequence information of the mitochondrial genes (COI,
Cyt b and 16S rRNA) can be used as a diagnostic molecular marker for identification and resolution of
taxonomic ambiguity in Ompok species.
