CARBOHYDRATE
INTRODUCTION
 Carbohydrate are the most abundant organic
molecules in nature.
 They are primarily composed of the elements
carbon, hydrogen, oxygen.
 The name carbohydrate literally means ‘hydrates
of carbon’
 Carbohydrate maybe defined as
polyhydroxyaldehyde or ketone or compounds
which produce them on hydrolysis.

FUNCTIONS OF CARBOHYDRATE
MONOSACCHARIDE
 Greek: mono- one
 They are the simplest group of
carbohydrate and are often referred
to as simple sugar.
 General formula Cn(H20)n
 Cannot be further hydrolyzed.
MONOSACCHARIDE
 Based on the functional group and the number of
carbon atoms-----further divide into
 Aldoses: aldehyde
 Ketoses: keto

 The monosaccharides are regarded as
Glucose
OLIGOSACCHARIDES
 Greek -Oligo- few
 Contain 2 to 10 monosaccharides molecules which are
liberated on hydrolysis.
 Based on the number of monosaccharides units present
the oligossacharides are divided into Disaccharides,
trisaccharides.
 Oligosaccharides with more than 3 monosaccharides are
not digested properly by human enzymes.
POLYSACCHARIDES
 Poly-many
 They are polymers of monosaccharide units with
high molecular weight.
 They are usually tasteless (non-sugar) and form
colloids with water.
 The polysaccharides are of two types- homo-
polysaccharide and hetro-polysaccharide

 Stereoisomerism is an important character of
monosaccharide.
 Stereoisomers are the compounds that have the same
structural formulae but differ in their spatial configuration.
 A carbon is said to be asymmetric when it is attached to
four different atoms or groups.
 The number of asymmetric carbon atom (n) determines
the possible isomers of a given compound which is equal
to 2n. Glucose contains 4 asymmetric carbons and thus
has 16 isomers.

OPTICAL ACTIVITY OF SUGARS
 Optical activity is a characteristic feature of compounds
with asymmetric carbon atom.
 When a beam of polarized light is passed through a
solution of an optical isomer, it will be rotated either to the
right or left.
 The term dextrorotatory (d+) and levorotatory (l-) are used
to compounds that respectively rotate the plane of
polarized light to the right or to the left.

REACTION OF MONOSACCHARIDE
A. Tautomerization or Enolization
The process of shifting a hydrogen atom from one carbon
atom another to produce enediols is known as
tautomerization. Sugars processing anomeric carbon atom
undergo tautomerization in alkaline solutions.
When glucose is kept in alkaline solution for several hours, it
undergoes isomerization to from D-fructose and D-
mannose.this reaction- known as the Lobry de Bruyn-von
Ekenstein transformation- results in the formation of a
common intermediate- namely enediol- for all the three
sugars.
The enediols are highly reactive hence sugars in alkaline
solution are powerful reducing agents.
REDUCING SUGAR
 The sugar are classified as reducing or non reducing.
The reducing property is attributed to the free aldehyde
or keto group of anomeric carbon.
 In the laboratory many tests are employed to identify the
reducing sugar. Benedicts test, fehling test, barfoed
test etc. the reduction is much more efficient in the
alkaline medium than in the acid medium.
 The sugars reduce cupric ions (cu2+)of copper sulphate
to cuprous ions (Cu+) which form a yellow precipitate of
cuprous hydroxide or a red precipitate of cuprous oxide

OXIDATION
 Depending on the oxidizing agent used, the terminal
aldehyde ( or keto) or the terminal alcohol or both the
groups maybe oxidized.
1. Oxidation of aldehyde group (CHO------COOH) results
in the formation of gluconic acid.
2. Oxidation of terminal alcohol group (CH2OH-----
COOH) leads to the production of glucuronic acid.
REDUCTION
 When treated with reducing agents such as sodium
amalgam, the aldehyde or keto group of
monosaccharides is reduced to corresponding alcohol.
DEHYDRATION
 When treated with conc sulfuric acid, monosaccharides
undergo dehydration with an elimination of 3 water
molecules.
 Thus hexoses give hydroxymethyl furfural while pentoses
give furfural on dehydration.
 These furfurals can condense with phenolic compounds (α
napthol) to form coloured products.
 This is the chemical basis of the popular Molisch test.
OSAZONE FORMATION
 Phenylhydrazine in acetic acid when boiled with
reducing sugar forms osazones.

DISACCHARIDES
 Disaccharides consists of two monosaccharide units held
together by a glycosidic bond. They are crystalline,
water soluble and sweet to taste. The disaccharides are
two types.
1. Reducing disaccharides with free aldehyde or keto
group e.g maltose, lactose.
2. Non reducing disaccharides with no free aldehyde or
keto group eg sucrose trehalose.
MALTOSE
 Maltose is composed of two α D-glucose units held
together by α(14) glycosidic bond.
 The free aldehyde group present on C1 of second glucose
answers the reducing reactions, besides the osazone
formations (sunflower-shaped). Maltose can be hydrolysed
by dilute acid or the enzyme maltase to librate two
molecules of α D-glucose.
 In isomaltose the glucose units are held together by α
(16) glycosidic linkage.
 Cellobiose is another disacchrides, identical in structure
with maltose, except that the former has β (14)
glycosidic linkage. Cellobiose is formed during the
hydrolysis of cellulose
SUCROSE
 Sucrose (cane sugar) is the sugar of commerce,
mostly produced by sugar cane and sugar beets.
Sucrose is made up to α D-glucose and β D-
fructose.
 The two monosaccharides are held together by a
glycosidic bond (α1β2) between C1 of α glucose
and C2 of β fructose. The reducing groups of
glucose and fructose are involved in glycosidic
bond hence sucrose is non reducing sugar and it
cannot form osazone.
SUCROSE
 Sucrose is an important source of dietary carbohydrate. It
is sweeter than most other common sugars (except
fructose) namely glucose, lactose and maltose. Sucrose
is employed as a sweetening agent in food industry.
INVERSION OF SUCROSE
 Sucrose as such is dextrorotatory (+66.5⸰
)
 But when hydrolyzed, sucrose becomes levorotatory (-28.2 ⸰
)
 The process of change in optical rotation from dextrorotatory (+) to
levorotatory (-) is referred to as inversion.
 The hydrolyzed mixture of sucrose, containing glucose and fructose,
is known as invert sugar.
 Hydrolysis of sucrose by the enzyme sucrase (invertase) or dilute acid
liberates one molecule each of glucose and fructose. It is postulated
that sucrose (dextro) is first spilt into α-D-glucopyranose (+52.5 ⸰
)
and β-D- fructofuranose both being dextrorotatory. However β-D-
fructofuranose is less stable and immediately gets converted to β-D-
fructofuranose which is strongly levorotatory (-92 ⸰
).
 The overall effect is that dextro sucrose (+66.5⸰
) on inversion is
converted to levo form (-28.2⸰
).

INTRODUCTION  Carbohydrate are the most abundant organic molecules in nature.  They are primarily composed of the elements carbon, hydrogen, oxygen.  The name carbohydrate literally means ‘hydrates of carbon’  Carbohydrate maybe defined as polyhydroxyaldehyde or ketone or compounds which produce them on hydrolysis.

MONOSACCHARIDE – STRUCTURAL ASPECT  Stereoisomerism is an important character of monosaccharide.  Stereoisomers are the compounds that have the same structural formulae but differ in their spatial configuration.  A carbon is said to be asymmetric when it is attached to four different atoms or groups.  The number of asymmetric carbon atom (n) determines the possible isomers of a given compound which is equal to 2n. Glucose contains 4 asymmetric carbons and thus has 16 isomers.

OPTICAL ACTIVITY OF SUGARS  Optical activity is a characteristic feature of compounds with asymmetric carbon atom.  When a beam of polarized light is passed through a solution of an optical isomer, it will be rotated either to the right or left.  The term dextrorotatory (d+) and levorotatory (l-) are used to compounds that respectively rotate the plane of polarized light to the right or to the left.  RACEMIC MIXTURE HOME WORK
  • 18. REACTION OF MONOSACCHARIDE A. Tautomerization or Enolization The process of shifting a hydrogen atom from one carbon atom another to produce enediols is known as tautomerization. Sugars processing anomeric carbon atom undergo tautomerization in alkaline solutions. When glucose is kept in alkaline solution for several hours, it undergoes isomerization to from D-fructose and D- mannose.this reaction- known as the Lobry de Bruyn-von Ekenstein transformation- results in the formation of a common intermediate- namely enediol- for all the three sugars. The enediols are highly reactive hence sugars in alkaline solution are powerful reducing agents.
  • 19. REDUCING SUGAR  The sugar are classified as reducing or non reducing. The reducing property is attributed to the free aldehyde or keto group of anomeric carbon.  In the laboratory many tests are employed to identify the reducing sugar. Benedicts test, fehling test, barfoed test etc. the reduction is much more efficient in the alkaline medium than in the acid medium.  The sugars reduce cupric ions (cu2+)of copper sulphate to cuprous ions (Cu+) which form a yellow precipitate of cuprous hydroxide or a red precipitate of cuprous oxide

  • 22. OXIDATION  Depending on the oxidizing agent used, the terminal aldehyde ( or keto) or the terminal alcohol or both the groups maybe oxidized. 1. Oxidation of aldehyde group (CHO------COOH) results in the formation of gluconic acid. 2. Oxidation of terminal alcohol group (CH2OH----- COOH) leads to the production of glucuronic acid.
  • 23. REDUCTION  When treated with reducing agents such as sodium amalgam, the aldehyde or keto group of monosaccharides is reduced to corresponding alcohol.

DEHYDRATION  When treated with conc sulfuric acid, monosaccharides undergo dehydration with an elimination of 3 water molecules.  Thus hexoses give hydroxymethyl furfural while pentoses give furfural on dehydration.  These furfurals can condense with phenolic compounds (α napthol) to form coloured products.  This is the chemical basis of the popular Molisch test.

  • 26. DISACCHARIDES  Disaccharides consists of two monosaccharide units held together by a glycosidic bond. They are crystalline, water soluble and sweet to taste. The disaccharides are two types. 1. Reducing disaccharides with free aldehyde or keto group e.g maltose, lactose. 2. Non reducing disaccharides with no free aldehyde or keto group eg sucrose trehalose.

MALTOSE  Maltose is composed of two α D-glucose units held together by α(14) glycosidic bond.  The free aldehyde group present on C1 of second glucose answers the reducing reactions, besides the osazone formations (sunflower-shaped). Maltose can be hydrolysed by dilute acid or the enzyme maltase to librate two molecules of α D-glucose.  In isomaltose the glucose units are held together by α (16) glycosidic linkage.  Cellobiose is another disacchrides, identical in structure with maltose, except that the former has β (14) glycosidic linkage. Cellobiose is formed during the hydrolysis of cellulose

  • 30. SUCROSE  Sucrose (cane sugar) is the sugar of commerce, mostly produced by sugar cane and sugar beets. Sucrose is made up to α D-glucose and β D- fructose.  The two monosaccharides are held together by a glycosidic bond (α1β2) between C1 of α glucose and C2 of β fructose. The reducing groups of glucose and fructose are involved in glycosidic bond hence sucrose is non reducing sugar and it cannot form osazone.
  • 31. SUCROSE  Sucrose is an important source of dietary carbohydrate. It is sweeter than most other common sugars (except fructose) namely glucose, lactose and maltose. Sucrose is employed as a sweetening agent in food industry.

INVERSION OF SUCROSE  Sucrose as such is dextrorotatory (+66.5⸰ )  But when hydrolyzed, sucrose becomes levorotatory (-28.2 ⸰ )  The process of change in optical rotation from dextrorotatory (+) to levorotatory (-) is referred to as inversion.  The hydrolyzed mixture of sucrose, containing glucose and fructose, is known as invert sugar.  Hydrolysis of sucrose by the enzyme sucrase (invertase) or dilute acid liberates one molecule each of glucose and fructose. It is postulated that sucrose (dextro) is first spilt into α-D-glucopyranose (+52.5 ⸰ ) and β-D- fructofuranose both being dextrorotatory. However β-D- fructofuranose is less stable and immediately gets converted to β-D- fructofuranose which is strongly levorotatory (-92 ⸰ ).  The overall effect is that dextro sucrose (+66.5⸰ ) on inversion is converted to levo form (-28.2⸰ ).
