1) Abscisic acid (ABA) induces stomatal closure in pea plants by raising both the cytosolic pH and nitric oxide (NO) levels in guard cells.
2) The rise in cytosolic pH occurs earlier than the increase in NO, suggesting that pH increases are upstream of NO production during ABA-induced stomatal closure.
3) Modulators that raise cytosolic pH like methylamine enhance stomatal closure and NO production by ABA, while agents that lower pH like butyrate prevent the effects of ABA.

ABSTRACT: Wild-type Pyrococcus furiosus is most likely unable to grow on chitin in the natural biotope due to a nucleotide insertion which separates the chitinase gene into two ORFs, whereas a genetically engineered strain with the deleted nucleotide is able to grow on chitin. In the latest studies, the recombinant enzyme activity against the crystal chitins was examined. But there are still some conflictions. In our study, to shed a light on whether the construct composed of a catalytic domain and a chitin binding domain show any activity against crystalline chitin, the construct was created in the pET 28b (+) expression vector and expressed in Escherichia coli. The chitinase with an approximately 55 kDa molecular weight was determined. The activity of the enzyme was measured spectrophotometrically. Despite the presence of enzyme activity against the colloidal chitin, no significant activity against the crystal chitin has been measured.

Abstract— Oxidative DNA damage is involved in the f cell death induced by freeze-dried powder during storage. Cell 8-hydroxy-2’deoxyguanosine (8-oxodG) is widely accepted as a biomarker of the “freeze-dried bacteria” oxidative DNA damage. The aim of this study was to introduce a method for determination 8-oxodG in cell freeze-dried samples using high-performance liquid chromatography with electrochemical detection. In the tested range of 0.5 µmol L-1 to 1.0 nmol L-1, the calibration curve was linear (r2=0.9995) and the limit of detection was 0.05 µmol L-1. The used method did not allow highlighting the presence in the samples of the 8OH within the limits of detection. A more successful method (more sensitive) would be needed to detect possibly the 8OH.

Low temperature is an important environmental factor that limits the survival, productivity and geographical distribution of plants. Oil seeds are the second global food resources among which Brassica napus L. is the third annual oil seed in the world. In cold stress, some biochemical and physiological reactions occur in response to reactive oxygen species (ROS). Hence, the effect of foliar application of salicylic acid (SA) on total chlorophyll content, malondialdehyde, and antioxidant enzymes activity and solute protein and proline contents were assessed in two canola varieties (Brassica napus L., cv RGS and LICORD) leaves exposed to cold stress during 0, 24, and 48 hours after salicylic acid treatment. They were first grown in a controlled growth room at 22/20 °C (day/night) for one month followed by SA spraying application (100, 200 and 400µM) and then plots were transferred to a cold environment (-2 °C) for 3 days. The results showed that the total chlorophyll content was decreased in RGS cultivars related to high salicylic acid concentration during the experiment. The results of antioxidant status showed that superoxide dismutase (SOD), peroxidase (POX), and also lipid peroxidation were increased significantly after 48 hours compared first day. Catalase (CAT) activity was decreased 24 hours after salicylic acid treatment. Results showed an increase in protein content in both cultivars treated with SA, by contrast proline was greatly affected by salicylic acid treatment and its content was the highest 24 hours after treatment. According to the results of the present study indicated that application of salicylic acid has useful effects on the biochemical traits. Thereupon it may be effective for the improvement of plant growth in cold regions.

Aim of the work: The present study was conducted to elucidate the possible protective effect of alpha lipoic acid (α-LA) against the deleterious effect perturbation induced in rat brain exposed to lead acetate. Methods: 32 Wistar male rats (weighing 130 ± 10 g) were divided into four groups (n=8): (1) normal control group (C); (2) Initiation group (Pb as lead acetate 20 mg/kg.b.wt, i.p. for 2 wks); (3) treatment group (α-LA 20 mg/kg.b.wt, i.p. for 3 wks); (4) post-initiation treatment group (Pb for 2 wks then followed by α-LA for 3 wks). Levels of monoamines (norepinephrine NE and dopamine DA), the level of Ache activity and finally adenosine triphosphate (ATP), were estimated in the hippocampus and cerebral cortex, in addition, a Morris water maze and the histological study were performed after completion of the experiments. Results: The results of the present work demonstrated that Pb inhibited neurotransmitters releases and decrease the level of Ache activity, as well as it inhibited energy production ATP. Pb impaired performance on Morris Water Maze of rats and histological degeneration. However, treatment with α-LA significantly attenuated the behavioral impairment and biochemical parameters in rat treated with Pb. And amelioration of histological changes. Conclusion: As a conclusion, treatment with α-LA can improve the Pb-induced toxicity via antioxidant activity.
