Cây phát sinh thu được theo phương pháp Bayesian dựa trên khung đọc gen matK
hoàn toàn tương đồng với cây phân loại thu được dựa theo phương pháp MP. Cụ thể, mẫu C4
cùng nhánh với mẫu C. kawakamii với PP là 100%, các mẫu còn lại nằm ở cùng một nhánh
với chỉ số của nhánh là 100%, trong nhóm tồn tại các dưới loài với chỉ số PP cao, lần lượt là:
nhánh C11, C12, C18 là 99%; C14, C15, C16, C17 là 74%, và của nhánh C2, C21 là 100%.
Điều này khẳng định lại một lần nữa, gen matK có khả năng phân tách tốt hơn vùng gen ITS,
có thể sử dụng gen này để xác định các dưới loài.
KẾT LUẬN VÀ KIẾN NGHỊ
Kết luận
Từ các kết quả thu được, chúng tôi đưa ra một số kết luận chính như sau:
1. Đã khuếch đại thành công hai vùng gen ITS và matK, và kết quả phân tích sự đa
hình trên mỗi vùng gen cho thấy vùng gen ITS kích thước từ 638-650 bp xuất hiện 113 điểm
đa hình, chiếm 17,4% trên toàn bộ trình tự vùng gen, có độ đa hình cao hơn so với gen matK
kích thước 871 bp với 61 điểm đa hình xuất hiện, chiếm 7% trên toàn bộ trình tự
2. Từ kết quả so sánh và phân tích trình tự ADN vùng gen ITS có thể xác định các
mẫu nghiên cứu C2, C11, C12, C13, C14, C15, C16, C17, C18, C20, C21 thuộc loài C.
javanica và mẫu C4 thuộc loài C. tangshen
3. Với sự tương đồng 100% trong trình tự, vùng gen ITS là rất bảo thủ ở loài C.
javanica và C. tangshen, rất có ý nghĩa trong kiểm định dược liệu.
4. Xây dựng cây phát sinh chủng loại dựa trên phương pháp Tiết kiệm tối đa
(Maximum Parsimony) và Bayesian dựa trên khung đọc riêng rẽ từng gen ITS và matK cho
kết quả tương đồng, cho thấy độ đáng tin cậy của cây thu được cũng như của 2 phương pháp
MP và Bayesian sử dụng để xây dựng cây chủng loại phát sinh trong chi Codonopsis. Các
cây phân loại xây dựng được một lần nữa cho kết luận xác định các mẫu nghiên cứu thuộc hai
