• Iron directly participates in the generation of toxic oxygen and
nitrogen intermediates.
• In circulation transferrin-Fe3+ complex.
• In cytosol ferritin chelates Fe 3+.
• Translocation of Fe 2+ is performed by the divalent cation
efflux pump Nramp2.

• Many bacteria internalize iron sequestered by siderophores.
• Macrophages counter attack by secreting lipocalin 2.
• Protozoa can not acquire Fe3+ thruogh siderophores and
siderophore receptors.
• Leishmania spends its intracellular life cycle inside the
endosomal pathway,where one of the major sources of iron is
Fe3+ complexed to transferrin,or Fe2+ generated from Fe3+
by endosomal reductases.
• Many bacteria internalize iron sequestered by siderophores.

• Macrophages generate an oxidative burst, which produces highly toxic
reactive oxygen intermediates including superoxide anion(O2-).
• Metalloenzyme superoxide dismutase (SOD) helps in detoxifing O2-,by
converting it into H2O2 and H2O.
• For SOD to work properly iron is must.
• Resistance to oxidative stress is correlated with the ability of Leishmania to
proliferate within macrophages.
• The endosomal pathway of macrophages is actively depleted in Fe 2+ by the
divalent cation efflux pump Nramp 1 and Nramp 2.
• Recent finding says L. Chagasi express a NADPH-dependent iron
reductase,capable of converting oxidized ,ferric into more soluble ferrus.

Leishmania amazonensis ferrous iron
transporter LIT 1
• LIT 1 ,the first ferrous iron transporter to be detected in trypanosomatid
protozoa.
• The 1 st gene encoding a Fe 2+ transporter to be cloned from plants or
animals was the Arabidopsis thaliana IRT 1.
• This encodes an integral membrane protein with eight predicted
transmembrane domains and potential metal binding sites rich in histidine
residues.
• Iron is preferred metal substrate for IRT 1,although cadmium, cobalt,
manganese and zinc can also be transported.
• LIT 1 ,the first ferrous iron transporter to be detected in trypanosomatid
protozoa.
