The present project deals with application of BUSALFAN as
haematopoiesis suppressive agent in inbred Swiss albino mouse. Functional
manifestation was carried out by observing the Peripheral Blood Haemogram.
Busalfan is a polyfunctional-alkylating agent, which can induce
haematopoietic failure that can be manifested by observing the peripheral blood
haemogram. Furthermore, by performing ‘single cell isolation technique’, the
splenocytes were isolated which contain lymphocytes, macrophages etc.
NitroBlueTetrazolium Assay was performed on these isolated splenocytes.
NitroBlueTetrazolium chloride is a double tetrazole structure, forming purple
formazone product, helping in the detection and approximation of phagocytic
ability of the splenocytes.
FACS (Fluorescence Activated Cell Sorter) analysis was
performed on the isolated splenocytes. It’s a latest flowcytometric technique, which
is very helpful in the observation of physical characteristics of the cells. The cell size
can be studied by the FACS analysis and even the granularity of the isolated
splenocytes can be studied by performing the FACS analysis of the isolated
splenocytes.
All the above-mentioned experiments were performed on the two
different groups of mouse. One was normal Swiss albino mouse and the other was
induced with busalfan for the comparative study.
