1. TECNOPHOR ELECTROPORATION
Nowadays, there is much of debate about the dynamics of cutaneous absorption and trans-
epidermal of various molecules, bearing in mind that, the main difficulty for the release of
substances through the human skin, is constituted by the outer layer, i.e. the stratum
corneum, … several theories have hypothesized...
Just to name a few:
1) one of the layers of the skin, represented by a row of cells in phase of dissolution, (lack of
nucleus), would be waterproof due to electrostatic forces to the polar molecules
2) the cell membrane, also called “housing”, surrounds each individual cell, and constitutes
the outer wall, with its formation made out mosaic lipid-protein( fats and proteins), it would be
able to disseminate the molecules, with greater coefficient of lipid distribution, with a
predominance of fat.
The transport of molecules through the human skin is of great interest for the transdermal
delivery, deep within the excellent barrier formed by our skin, molecules of various kinds
useful to the skin and the possibility to carry drugs with in a totally non-invasive method.
Small lipid soluble molecules can be absorbed by the “stratum corneum”, and then diffuse
through the lipid double layer of cell membrane, but water-soluble molecules, can not
penetrate significantly through this barrier.
On the basis of the function of the K / Na pump, some authors from the 900' to date, have
searched for a similar mechanism for penetration of water molecules.
The existence of canals that continuously mediate the flow of water molecules and small
solutions through the biological tissues, as well as through the wall of the urinary tract, or
even through the membrane of individual cells, was hypothesized in 1900.
Later in 1950 was discovered that water is rapidly transported through the cell membrane of
red blood cells, through selective canals for water that exclude ions or other solutes.
A more specific assumption is that short segments of canals are formed through the lipids of
the membrane in a double row of 5-6 cells comeociti, contiguous downwards, to create a trans-
cellular passage that we can precisely define "canel watery " suitable to the passage of water
molecules.
2. The story of why building a machine such as TECNOPHOR, able to create practically this
physical phenomenon known as "Electroporation", takes into consideration the study of a
preceding physical phenomena, made with other machines such as:
• Iontophoresis which creates the ionization of a substance through a continuous
galvanic current
• Iontophoresis as above, using however a evolved modulated series of wave
pulses.
The limitations of these techniques are:
• A low amount of active ingredient practically unable to convey
• Limited depth of tissue penetration
• Low concentration of active principle in an specific area
Electroporation is a transdermal delivery system, which overcomes the limitations of
passed techniques, and is used in medical therapy to convey drugs deeper into the skin
(by going up to 15 cm more in depth).
When an electrical pulse affects a cell, it generates a potential trans-membrane and
consequently the lipid layer of the cell membrane alters and gives rise to the formation
of "aqueous vessels", also called electro-pores.
These canals remain for a period of time, which generally runs from a few seconds to
about ten minutes. Generating " hydrophilic macro-pores" within the lipid bilayer of
the cell membrane, which seems to measure several microns in diameter, allowing the
passage of large molecules and higher molecular weight.
This phenomenon, which relates to the cell membrane is very important for the
progress of biology, biotechnology, and medicine, which allows the penetration of
active substances and water-soluble high molecular weight, such as for example,
hyaluronic acid, collagen, vitamin C, etc... by introducing up to 90% of the active
substance.
In practice, with this method we can convey water-soluble molecules through the
epidermal barrier, which barrier has a very low permeability to water-soluble
substances.
The electroporation device, Tecnophor, consists of an ionization chamber in the hand-
piece, capable of ionizing the molecules, and thus render "active", the transport of any
water-soluble substance.
Then through the opening of the electro-pores on the skin, the substance pass through
osmosis from one cell to another, and are released in the deeper transdermal skin
compartment.
3. The hand-piece in contact with the skin, incorporates a dispenser containing
conductive gel in which are dispersed the active ingredients that need to be conveyed

4. The Skin

permeability of tissues. This occurs, when a cell through an electrical pulse, at selected
frequencies, and generates a potential trans-membrane between minimum 0.5 and 1.5
volts; the lipid bilayer of the cell is subjected to a temporary readjustment, with the
formation of aqueous vessels in the cell membrane, called electro-pores.
Such electro-pores determine the passage of molecules which otherwise would not
pass the stratum corneum.
All this without suffering, no scalpel and without the use of needles or stings, acting
directly on cellular mechanisms without creating any side effects.

6. What happens during electroporation?
