This document discusses why 3D cell biology is more difficult than 2D cell biology. In 2D cell cultures using standard live-cell imaging techniques, it is easier to get an instant visual assessment of whether cells are healthy and developing properly by looking at metrics like confluency, growth, and cell spreading - but these visual cues are not available in 3D cell cultures which do not allow the same type of overhead viewing as a 2D monolayer.
