Mental health has been in the news quite a bit lately. Dozens of U.S. states are currently suing Meta for contributing to the youth mental health crisis by inserting addictive features into their products, while the U.S. Surgeon General is touring the nation to bring awareness to the growing epidemic of loneliness and isolation. The country has endured periods of low national morale, such as in the 1970s when high inflation and the energy crisis worsened public sentiment following the Vietnam War. The current mood, however, feels different. Gallup recently reported that national mental health is at an all-time low, with few bright spots to lift spirits.
To better understand how Americans are feeling and their attitudes towards mental health in general, ThinkNow conducted a nationally representative quantitative survey of 1,500 respondents and found some interesting differences among ethnic, age and gender groups.
Technology
For example, 52% agree that technology and social media have a negative impact on mental health, but when broken out by race, 61% of Whites felt technology had a negative effect, and only 48% of Hispanics thought it did.
While technology has helped us keep in touch with friends and family in faraway places, it appears to have degraded our ability to connect in person. Staying connected online is a double-edged sword since the same news feed that brings us pictures of the grandkids and fluffy kittens also feeds us news about the wars in Israel and Ukraine, the dysfunction in Washington, the latest mass shooting and the climate crisis.
Hispanics may have a built-in defense against the isolation technology breeds, owing to their large, multigenerational households, strong social support systems, and tendency to use social media to stay connected with relatives abroad.
Age and Gender
When asked how individuals rate their mental health, men rate it higher than women by 11 percentage points, and Baby Boomers rank it highest at 83%, saying it’s good or excellent vs. 57% of Gen Z saying the same.
Gen Z spends the most amount of time on social media, so the notion that social media negatively affects mental health appears to be correlated. Unfortunately, Gen Z is also the generation that’s least comfortable discussing mental health concerns with healthcare professionals. Only 40% of them state they’re comfortable discussing their issues with a professional compared to 60% of Millennials and 65% of Boomers.
Race Affects Attitudes
As seen in previous research conducted by ThinkNow, Asian Americans lag other groups when it comes to awareness of mental health issues. Twenty-four percent of Asian Americans believe that having a mental health issue is a sign of weakness compared to the 16% average for all groups. Asians are also considerably less likely to be aware of mental health services in their communities (42% vs. 55%) and most likely to seek out information on social media (51% vs. 35%).

oporności na powszechnie stosowane antybiotyki
występującej wśród bakterii środowiskowych
wyizolowanych z różnych gleb woj. świętokrzyskiego.
Jakie to antybiotyki i dlaczego?
W badaniach stosuję 7 antybiotyków ß-laktamowych.
Są to antybiotyki, które nie pozwalają na tworzenie
się ściany komórek bakterii- co uniemożliwia
jednocześnie tworzenie się miliardów nowych
komórek. Preparaty te są bardzo często stosowane w
leczeniu zakażeń powodowanych przez bakterie.
Szczepy z jakich gleb badam?
Do badań wykorzystałam 12 prób różnych gleb
(kwaśnych, zasadowych, zanieczyszczonych
związkami toksycznymi np. metalami ciężkimi). Z
tego materiału wyhodowałam i wyizolowałam różne
bakterie.
4. Usytuowanie miejsc z których zostały pobrane próbki gleb, z których
następnie wyizolowano szczepy użyte w badaniach.

glebowym: 1 – 5 – gleby kwaśne z okolic
kopalni siarki (okolice Tarnobrzega); 6 – gleba
uprawna z terenów czystych ekologicznie
(okolice Klonowa); 7 i 8 – gleba skażona
metalami ciężkimi (Kielce); 9 i 11 – gleba
zasadowa (okolice Nowin pod Kielcami);
10 – gleba z terenów zurbanizowanych (Kielce,
centrum); 12 – gleba skażona uranem (Rudki).
5. DLACZEGO AKURAT GLEBA I DLACZEGO
SZUKAM W NIEJ BAKTERII OPORNYCH ?
Gleba jest najbardziej sprzyjającym środowiskiem
do życia bakterii. Jest w niej dużo składników
odżywczych i wilgoci dzięki czemu
mikroorganizmy dobrze się w niej rozwijają. Gleba
chroni również bytujące w niej bakterie przed
szkodliwym promieniowaniem
UV. Dlatego ilość bakterii, jaka
w niej żyje jest olbrzymia. Część
z nich (ok. 1 do 4%) potrafimy
wyhodować w laboratorium.

6. W glebie występują różne drobnoustroje produkujące
antybiotyki, np. grzyby, z których pozyskano penicylinę
– pierwszy antybiotyk ß-laktamowy. Często trafiają tam
również inne związki przeciwbakteryjne. Jednym z ich
podstawowych źródeł są nawozy naturalne pochodzące
z gospodarstw lub pozostałości z oczyszczalni ścieków.
Powszechnie stosowaną praktyką w hodowli zwierząt
jest dodawanie do pasz
antybiotyków
chroniących zwierzęta
przed epidemiami. Zaś
w ściekach trafiających
do oczyszczalni
znajdują się związki i
antybiotyki, które nie
zostały wykorzystane

funkcjonuje polityka terapii
antybiotykami, która
określa jakie antybiotyki
można używać i umożliwia
monitorowanie narastania

Z tych powodów gleba
może być rezerwuarem

8. W moich badaniach poszukiwałam bakterii wykazujących kilka
określonych typów oporności:
• ESBL – jest to oporność na
większość (ok. 84%)
antybiotyków ß-laktamowych.
Bakterie wykazujące taką
oporność można zabić tylko
ok.16% tych antybiotyków.
• MBL – zazwyczaj niemal
wszystkie (prawie 98%)
antybiotyki ß-laktamowe nie
są skuteczne wobec bakterii
wykazujących ten typ
oporności.
• KPC – bakterie wykazujące
ten typ oporności są zwykle
niewrażliwe na ok. 62% http://www.news-

antybiotyków ß-laktamowych.
9. A dlaczego akurat taka oporność jest ważna?
Zarówno oporność typu ESBL, MBL jak i KPC występuje na elementach
genetycznych, które nie są ściśle związane z określoną bakterią. Mogą się one
stosunkowo łatwo rozprzestrzeniać a bakterie mogą przekazywać je sobie
nawzajem. W rezultacie gwałtownie wzrasta ilość bakterii opornych.

Określenie występowania oporności na ampicylinę – jeden
z najczęściej i najdłużej stosowanych antybiotyków
ß-laktamowych.

Ten etap badań polegał na
hodowli wyizolowanych bakteria oporna
szczepów bakterii na podłożu
mikrobiologicznym z
dodatkiem ampicyliny. Na
takiej pożywce wyrastały
jedynie szczepy oporne.
Jednocześnie był to wstępny
skrining, ponieważ bakterie
wykazujące interesujące mnie
typy oporności (czyli
ESBL, MBL i KPC) są również
oporne na ampicylinę.

13. II ETAP BADAŃ
Określenie występowania oporności typu ESBL.
Do tego realizacji tego etapu badań stosowałam test z wykorzystaniem krążków
nasączonych odpowiednimi antybiotykami. Krążki te układałam w ściśle określony
sposób na płytce z pożywką, na której uprzednio rozprowadziłam zawiesinę bakterii.
Bakteria była klasyfikowana jako ESBL jeśli widziałam charakterystyczne
niesymetryczne strefy wokół określonych krążków z antybiotykami.

Wykonywałam test z wykorzystaniem krążka nasączonego odpowiednim
antybiotykiem oraz krążka z antybiotykiem i substancją hamującą oporność MBL.
Układałam je w określony sposób na płytce z uprzednio rozprowadzoną zawiesinę
bakterii.
Bakteria była klasyfikowana jako MBL jeśli różnica pomiędzy strefami zahamowania
wzrostu bakterii była równa lub większa niż 5 mm.

18. Identyfikacja bakterii, czyli określenie ich gatunku, które
wykazują oporność typu ESBL, MBL lub KPC.
Jest to o tyle ważne, bo taka oporność jest badana tylko dla kilku ściśle
określonych gatunków bakterii, zwykle związanych z człowiekiem. Jest
bardzo prawdopodobne, że oporność którą wykryłam występuje u innych
gatunków niż już opisywane. Niestety bakterii środowiskowych nie można
zidentyfikować powszechnie dostępnymi testami komercyjnymi. Trzeba
wykorzystywać metody genetyczne, które są znacznie bardziej
skomplikowane i kosztowne.
Określenie, czy oporność którą wykrywałam jest
warunkowana przez znane i opisane już geny.
W naukowych publikacjach jest opisanych kilka genetycznych metod
weryfikacji typów oporności bakterii. Jeśli żadna z nich nie będzie
skuteczna wobec moich bakterii, to oznaczać to będzie, że znalazłam
nowe, nieznane wcześniej geny warunkujące antybiotykoodporność.
19. Określenie nowych genów warunkujących oporność, w
szczególności MBL lub KPC.
Jeśli okaże się, że któryś z moich szczepów bakterii wykazuje oporność wcześniej
nie opisywaną to będę mogła określić jak wygląda gen, który ją warunkuje. Niestety
wymaga to stosowania metod inżynierii genetycznej, które też do najtańszych nie
należą.
Instytucja, w której prowadzę moje
badania posiada aparaturę, która
umożliwi mi realizację tych
planów. Mam do nich wszystkich
dostęp, jak również wsparcie
innych pracowników. Niestety
jednostka ta nie jest w stanie
sfinansować kosztownych
odczynników, które są mi
niezbędne do osiągnięcia
przedstawionych powyżej celów.
