Occam’s razor prompts us to simplify explanations, and this talk will present how a very simple transform has been leveraged to improve performance of both generative and discriminative learners, as well as unsupervised learning, in a number of application domains including differentially private community discovery.

Sarcasm and puns aside, you’ve got a point. It isn’t easy to switch methodologies in the middle of a project. Doc shares some stories from his own experiences helping teams make this change and provides a few pointers that can help you do the same.

Scientific Research
I have a long standing research interest in cardiovascular epigenetics. Since I am at Goethe University
(May 2012) my main research activity has been focused on human subjects and specifically on the
characterization of epigenetic mechanisms important for the homeostasis of the human vascular and
cardiac districts with special attention paid to pathophysiological conditions that may affect cellular
and organ function including dysmetabolism and diabetes. My laboratory is currently focused on the
understanding of molecular cues at the basis of epigenetic memory and metabolic alterations in
human patients undergoing major cardiac surgery. Special attention is given to the metabolomics
profile of those metabolites important for epigenetic processes including DNA methylation and
subsequent modification of methylated cytosines including formation of hydroxymethyl‐, formyl‐ and
carboxyl‐cytosine. These modifications are currently determined in cardiac biopsis, in isolated cardiac
auricles, in isolated primary cardiac fibroblasts and in peripheral blood samples. In addition, I started
the characterization of a number of new epigenetically active molecules targeting epigenetic enzymes
important for DNA methylation control including DNA methyl transferases and DNA demethylases
alias Ten‐Eleven‐Translocation proteins (TETs). Specific aim of this research activity is to understand
the potential application of novel small molecule pro‐drugs to relevant human cardiovascular diseases
and cardiovascular risk factors including metabolic syndrome and diabetes. In brief, human cardiac
fibroblasts may be considered important to evaluate the effect metabolic alterations potentially
affecting adult cardiac stem cell function. Since, I started my current appointment at Goethe
University (May 2012) a number of human primary cardiac fibroblast preparations (about 50) have
been generated in close collaboration with Prof. Andreas M. Zeiher (Director of the Dept. of
Cardiology at Goethe University Hospital) and Prof. Andres Beiras‐Fernandez of the Thoracic Surgery
Clinic of the Goethe University Hospital. These cells have been used for investigations aimed at
understanding whether an altered metabolism, possible consequence of diabetes, may subsist as a cell
autonomous defect realizing a condition able to permanently alter the intracellular environment,
preventing or reducing the proliferation and differentiation potential of these cells. This research is
still in progress, however, our manuscript published in Diabetes on January 2014, indicates the
presence of horizontally inherited epigenetic defects in human cardiac stromal cells leading to the
accumulation of abnormal DNA methylation in cell obtained from patients with clinical history of
Diabetes. These alterations are detectable ex vivo and in well controlled metabolic conditions. Our
study is now focused on the results of an integrated metabolomic, transcriptomic, genomic approach
in order to understand at which level the alteration is most evident and at what extent. Parallel to this
research, in collaboration with other Institutions in Europe and US we started to characterize new
small molecules targeted to specific epigenetic enzymes deputed at DNA methylation regulation
including DNA methyl‐trasferases (DNMTs) and Ten‐eleven‐translocation (TET) enzymes. This
research recently led to the identification of new lead compounds capable of modulate the content of
methylated DNA in cardiac stromal cells rescuing, at least partially, the intrinsic functional defect. In
addition, in collaboration with Prof. Evelyn Ullrich of the Pediatric Oncology Clinic at KGU, I started a
to evaluate the effect of epigenetically active drugs on the proliferation of human Natural Killer cells
suited for antitumor activity. This activity is currently ongoing.

Specific Results:
Identification of the histone acetylase activator SPV106 as a potential activator of cellular
proliferation which has been now reported by us as effective on metabolically compromised
cardiac mesenchymal cell isolated from diabetic patients restoring function (1).
Evaluation of SPV106 effect of human natural killer cells proliferation and request for patent
regarding the use of SPV106 for the activation of primary human cell functions have been
approved by Goethe University. The final patent application is now submitted to the
European Patent Office (Application N. 14 179 989.0).
Characterization of new epigenetically active molecules, targeting DNA methylases and
demethylases (TET proteins). 5 compounds selected for further investigation. One of these
molecules received a first positive evaluation for the patent attorney and it is currently
preparing for a second evaluation by the Goethe University Patenting Committee in 2015 (see
attached document).
Isolation and characterization of 50 cardiac human fibroblast primary cultures obtained from
diabetic and non‐diabetic patients undergoing cardiac surgery.
