6. Statistical data of Plastic production
In Bangladesh
1.4 Lacs tons of PET resins is imported every year
400 crores of bottle produced per year
Plastic consumption 5 kg/person/year
Ref: Daily Star, 2016
7. PET
(Polyethelene terephthalate)
• Versatile use
• Low cost
• Stable
• Most abundant Plastic
• Easily Processable
It’s non-biodegradable property becomes
a great environmental threat
10. MECHANISM OF ‘PETase & MHETase’
Bacteria adhere to
PET
PETase secreted
PETase breaks PET into MHET
MHET uptaken by Bacteria
MHETase breaks MHET into
TPA & EG
TPA used as a carbon source
*PET = Polyethylene Terephthalate
*MHET = Mono-(2-hydroxyethyl) terephthalate
*TPA = Terephthalic acid
*EG = Ethylene Glycol
These Enzymes Activity:
pH range
15-42 (optimally at pH 7-7.5)
Temperature range
15-42 ºC (optimally at 30-37 ºC)
11. Recombination of Bacterial DNA
Isolation of PETase, MHETase &
associated transporter genes from the
Bacterial strain Ideonella sakaiensis 201 F6
Site directed mutations to get more active
enzyme
Transfer of genes to the viable bacteria
from plastic landfill of our country
Transferring of the genes will be done by
Biotechnology of DNA recombination.
12. Finding Out The Viable Bacteria
Adhere to PET
Colonize on the surface of
PET
Express the gene
Take up the gene of PETase
May use TPA as carbon
source for nutrition
Transport PETase outside
The Bacteria
with the
properties
13. Implementation
We will implement the bacteria to the landfill. After confirming-
Adequate proliferation and growth of the bacterial cell even after the DNA recombination
The transgenic bacteria can fulfill it’s nutritional requirement from the landfill
14. FUTURE GOAL
Reducing the implementation cost
Further research to enhance the activity of enzymes
Searching for other enzymes that can degrade other plastics
Involvement of Government & other organization to implement this in large
scale
Increasing public awareness
Planning a scheme for over all sustainable waste management program
