Teflabs announces new families of sodium and calcium dye indicators called Asante dyes. Asante Calcium Red is a ratiometric calcium indicator that is visible and excitable at 488 nm. Confocal imaging of rat cardiac myocytes loaded with Asante Calcium Red shows the dye reporting calcium transients during electric field stimulation. Testing was provided by researchers at the University of Maryland who showed the dye accurately reporting calcium levels. Asante Natrium v.1 is a visible sodium indicator with excitation at 500 nm and a dissociation constant of around 7 mM sodium.

8. Loading of rat cardiac myocytes
Acutely dissociated rat cardiac myocytes were incubated for 45 min at room
temperature with Tyrode’s solution containing 10 µM Asante Calcium Red
AM and 0.02% (wt/vol) Pluronic F-127. The cells were then transferred to
fresh Tyrode’s solution and allowed to stand for an additional 45 min to
ensure complete hydrolysis of the AM ester.

9. Confocal fluorescence imaging
Myocytes loaded with Asante Calcium Red were superfused with Tyrode’
solution and imaged on a Zeiss 5 Live laser scanning confocal microscope.
With 488-nm excitation, fluorescence images were acquired at 525 nm and
650 nm at 19 frames per second. The background fluorescence for each
image was defined as the mean intensity value in a region-of-interest located
near the cell but outside the cell contour. After background correction and
smoothing, at each time point, the 650-nm image was divided by the 525-
nm image to generate the ratio image. To evoke a Ca 2+ transient and
contraction, the cell was stimulated with an electric field pulse delivered
through platinum electrodes immersed in the Tyrode’s solution.
