2. “ENHANCEMENT OF SOLUBILITY & DISSOLUTION RATE OF ARIPIPRAZOLE BY
SOLID DISPERSION TECHNIQUE USING FUSION METHOD.”
The present study describes the preparation of solid dispersions designed to increase the
solubility and dissolution rate of Aripiprazole (APZ), an antipsychotic drug used in the
treatment of schizophrenia, bipolar disorder.
Solid dispersions of APZ were prepared by fusion method using water soluble carriers
like, polyethylene glycol 4000 & 6000 (PEG 4000&6000) and a mixture of PEG
4000,6000 with either CCS or CP and characterized by Fourier transform infrared
spectroscopy, differential scanning calorimetry and in vitro dissolution.
Physical mixtures containing APZ and PEG 4000 and PEG 6000 showed enhanced
dissolution of APZ as compared with pure drug, which indicates the solubilizing effect of
PEG. Dissolution results indicated a significant improvement in dissolution rate of APZ
from its solid dispersions as compared with Physical mixtures and that of pure drug.
Ternary solid dispersions were more effective to improve the dissolution rate of poorly
soluble drug than binary solid dispersions, physical mixtures and pure drug. Aqueous
solubility studies indicated that solubility of solid dispersions was increased when
compared with pure drug. FTIR and DSC studies suggesting that there was no physical or
chemical interaction in between APZ and carriers. From the in vitro dissolution studies,
tablets made from PEG 6000 the formulation 6F3 showed fast dissolution than 4F3
formulations.
