1. Cilj fiksacije
 očuvanje organa, tkiva i stanica u što prirodnijem obliku - „life-like state“

 očuvanje i zaštita kadavera doniranih u znanstvene i edukativne svrhe

Kada bi postojao idealan fiksativ trebao bi imati određena svojstva:
 Da ne očvršćuje tkivo, organe ili tijelo
 Da omogućuje savršeno bojanje tkiva za analize
 Da ne bude toksičan za ljude i životinje
 Da mu cijena bude prihvatljiva
 Idealan fiksativ NE postoji
3. Postupak fiksacije
 Uporaba samo formaldehida kao fiksativa ne daje dobre rezultate dugoročno
 Kombinacija formaldehida s drugim agensima
 Specijalizirani protokoli za fiksaciju KADAVERA ili samo izoliranog ORGANA
KAISERLINGOV PROTOKOL
• Za očuvanje pojedinih izoliranih organa u svrhu nastave
• Prednost ovog protokola je očuvanje približne boje tkiva
Priprema otopina:
 Kaiserling I: 85g kalijeva acetata, 45g kalijeva nitrata, 4800mL formalina (3 - 4%)
 Kaiserling II: etilni alkohol, 80 – 95%
 Kaiserling III: 200g kalijeva acetata, 300mL glicerina, 900mL vodovodne vode
PROTOKOL PO PRETORIUSU
 Standardizirana formula za fiksacija kadavera u svrhu nastave
 Prednost ovog protokola je trajnost kadavera i vrlo niska cijena provođenja fiksacije
 Nedostatak - smećkasto obojenje tkiva
 Priprema fiksativa:
• 96% etanol
• Formalin (3 – 4%) 500 mL
• Vovododna voda 750 mL
• 80% tekući fenol 3,5 L
Postupak injiciranja fiksativa:
• injiciranje pod tlakom od 1 do 1.5 kPa u tijelo
(kroz femoralnu arteriju) oko 5 L formalina
kadaver 60 kg → 25 L fiksativa → fiksacija traje 3. - 5. godina
 Održavanje kadavera
• Prekrivanje tkaninom umočenom u humektant te čuvanje u hladnoj prostoriji
 Priprema humektanta:
• Glicerin – 250 mL
• 80 % tekući fenol – 250 mL
• Vodovodna voda – do 1 L
