Answer : From the image the Fluroscent staining for the nucleie with DAPI staining is sucessful where as the staining with Phyllodin is not sucessfull as this has not stained the Actin filaments pressent around the nulceus. The dye Phyllodin stains the F-Actin filaments present around the Nucleus. It has stained the flagellum of a bacteria present in the endothelial smear. From the Phyllodin stinaining we are expected to see the actin filaments stained in green colour. The actin filaments were not visible in the current preparation Yes the nuclei are visible as Macro Nucleus and Micronucleus and were stained in Blue color. the factors may looked in for better preparation of the epithelial smear are as follows 1. Use freshly prepared fluroscent dye Phyllodin and DAPI and try different concentrations of the fluroscent dyes. 2. Prepartion of the smear needs to standardised 3. Try seperate staining for Phyllodin staining and and DAPI staining.
