Description
RBC Taq DNA Polymerase Mastermix is a ready-to-use mastermix. Convenient for amplification of DNA fragments without the
need to thaw individual components, reducing the risk of contamination and pipetting errors. RBC Taq DNA Polymerase
Mastermix utilises a Mg2+ base reaction buffer system with high quality RBC Taq DNA polymerase, containing all the nucleotides
and reagents necessary to perform a standard reaction.
Storage Buffer
RBC Taq DNA Polymerase Mastermix must be stored at either -20˚C or 4˚C. Storage at 4˚C avoids the necessity of thawing the mix
before assembling the reaction. For long term storage, -20˚C is recommended.
Component
Each vial contains 500μl of a 2X working concentration master mix. The addition of template and primers results in a 50ul final
reaction volume, containing
10 mM KCl, 1.5mM MgCl2, 20 mM Tris-HCl (pH8.8), 10 mM (NH4)2SO4, 0.1 mg/ml BSA, 0.2 mM dNTP mix, 1.25 U RBC Taq DNA
polymerase.
User may aliquot into microtubes according to desired reaction volume or according to recommendations below.

2. General Reaction Conditions
The following protocol is suggested as a starting point and guideline when using RBC Taq DNA Polymerase
Mastermix. We recommend assembling reactions on ice from pre-chilled components. This protocol is for a
reaction size of approximately 50 μl. The reaction size may be adjusted as desired.

Set up reaction tubes / plates on ice.

Add the following components in any order to each reaction vessel.
* 25μl for RBC Taq DNA Polymerase Mastermix
* Primers (200 nM final concentration per primer is recommended)
* Template DNA solution
3 Mix contents and run amplification program. Utilise standard amplification parameters for amplification of
vast majority of DNA sequences. User may also optimize for specific gene sequences.
