ÖZET
Kanser dünyada ölüm sebepleri arasında ilk sırada yer almaktadır. Klasik tedavi
yöntemlerine alternatif olarak kullanılan ‘PDT’ ana ilkesi ışığa duyarlı bir maddenin (PS)
uygun dalga boyundaki görünür ışığa maruz bırakılmasıyla oluşan serbest radikaller ve singlet
oksijenin (1
O2) yağ, protein ve nükleik asitler gibi birçok biyolojik molekülle etkileşip kanser
hücrelerini tahrip etmesidir. Kullanılan ışığa duyarlı maddenin toksisite oluşturmadan seçici
olarak tümör dokusunda birikmesi, ışığın sadece tümör bölgesine uygulanması ile normal
doku hasarının engellenmesi, istenilen sonuç elde edilene kadar tekrarlanabilir olması ve ucuz
maliyeti ile PDT geleneksel onkoterapi yöntemlerine göre bir avantaj sağlamaktadır.
Hasta popülasyonlarında tümör heterojenitesi fazla olabilir ve standart tedavi yöntemleri
uygulandığında alınan sonuçlar farklılık gösterebilmektedir. Teranostik molekülleri, tanı ve
tedaviyi aynı platformda birleştirebilir olması sebebiyle hastadan hastaya farklı uygulanabilir
tedavileri yani ‘kişiselleştirilmiş tedavi’yi mümkün kılmaktadır.
Bu tez çalışmasında tedavi kısmında PDT’nin , tanı kısmında ise; ‘click chemistry’
kullanılarak ftalosiyanin moleküllerine konjuge edilen MRG kontrast ajanı Gd-DOTA
kompleksleri kullanılması hedeflenmiştir. Bu amaçla sentezlenen metalli ftalosiyanin
molekülleri (M: Zn ve Ga) oligoetilen grupları ile sübsititüe edilerek sudaki çözünürlüklerinin
arttırılması hedeflenmiştir. Bu çerçevede sentezlenen ftalosiyanin moleküllerinin uygun
metotlarla saflaştırılmasının ardından, karakterizasyonu FT-IR, UV-Vis, Kütle (MS), 1
H-
NMR, 13
C-NMR spektrumları yardımları ile aydınlatılmıştır. Bu moleküllerin PDT ‘de
kullanılabilirliklerini araştırabilmek için fotofiziksel ve fotokimyasal özellikleri incelenmiştir.

SUMMARY
Cancer is among the leading causes of death worldwide. PDT is a promising alternative
to traditional treatments such as surgery, chemotherapy and radiotherapy. PDT utilizes a light-
sensitive material (PS) that forms free radicals and singlet oxygen (1
O2) upon exposure to
visible light of the appropriate wavelength. Singlet oxygen (1
O2) has the ability to destroy
cancer cells by damaging their biological molecules such as proteins, nucleic acids and lipids.
PDT has various advantages over traditional oncotherapy methods; i) light-sensitive materials
can be concentrated selectively in the tumor tissue without causing toxicity, ii) causes
minimal damage to healthy tissue by exposure of light only to the tumor tissue, iii) can be
repeated until obtaining the desired benefit, iv) low cost.
Tumour heterogeneity in patient populations can be vast and standard treatments can
lead to significant differences in patient outcomes. Personalized medicine is possible through
theranostics due to the combination of diagnostics and therapy in a single platform, allowing
optimization and personalization of the treatment.
In this thesis, our aim was to use phthalocyanine molecules both as photosensitizers for
PDT and as MRI contrast agents for diagnosis purposes through Gd-DOTA complexes,
covalently bonded to phthalocyanine macrocycle via ‘click chemistry’. In this regard,
synthesized phthalocyanine molecules (M: Zn, Ga) were substituted with oligoethylene
groups to achieve better solubility. Novel phthalocyanine molecules in this work were
purified by suitable means and characterized by FT-IR, UV-Vis, Mass (MS), 1
H-NMR and
13
C-NMR spectroscopy techniques. Photophysical and photochemical properties of
phthalocyanines were investigated to determine their performance in PDT applications.
