TtSmtB acts as a transcriptional repressor of an operon involved in arsenic resistance in the thermophilic bacterium Thermus thermophilus. The study characterized the role of TtSmtB by analyzing its interaction with target promoters both in vivo and in vitro. Results showed that TtSmtB directly binds to three arsenic-responsive promoters upstream of genes in the operon. Expression of these genes was deregulated in a T. thermophilus mutant strain lacking TtSmtB, confirming its role as a transcriptional repressor during arsenic stress response.
