Mental health has been in the news quite a bit lately. Dozens of U.S. states are currently suing Meta for contributing to the youth mental health crisis by inserting addictive features into their products, while the U.S. Surgeon General is touring the nation to bring awareness to the growing epidemic of loneliness and isolation. The country has endured periods of low national morale, such as in the 1970s when high inflation and the energy crisis worsened public sentiment following the Vietnam War. The current mood, however, feels different. Gallup recently reported that national mental health is at an all-time low, with few bright spots to lift spirits.
To better understand how Americans are feeling and their attitudes towards mental health in general, ThinkNow conducted a nationally representative quantitative survey of 1,500 respondents and found some interesting differences among ethnic, age and gender groups.
Technology
For example, 52% agree that technology and social media have a negative impact on mental health, but when broken out by race, 61% of Whites felt technology had a negative effect, and only 48% of Hispanics thought it did.
While technology has helped us keep in touch with friends and family in faraway places, it appears to have degraded our ability to connect in person. Staying connected online is a double-edged sword since the same news feed that brings us pictures of the grandkids and fluffy kittens also feeds us news about the wars in Israel and Ukraine, the dysfunction in Washington, the latest mass shooting and the climate crisis.
Hispanics may have a built-in defense against the isolation technology breeds, owing to their large, multigenerational households, strong social support systems, and tendency to use social media to stay connected with relatives abroad.
Age and Gender
When asked how individuals rate their mental health, men rate it higher than women by 11 percentage points, and Baby Boomers rank it highest at 83%, saying it’s good or excellent vs. 57% of Gen Z saying the same.
Gen Z spends the most amount of time on social media, so the notion that social media negatively affects mental health appears to be correlated. Unfortunately, Gen Z is also the generation that’s least comfortable discussing mental health concerns with healthcare professionals. Only 40% of them state they’re comfortable discussing their issues with a professional compared to 60% of Millennials and 65% of Boomers.
Race Affects Attitudes
As seen in previous research conducted by ThinkNow, Asian Americans lag other groups when it comes to awareness of mental health issues. Twenty-four percent of Asian Americans believe that having a mental health issue is a sign of weakness compared to the 16% average for all groups. Asians are also considerably less likely to be aware of mental health services in their communities (42% vs. 55%) and most likely to seek out information on social media (51% vs. 35%).

- Watertransportmaatschappij Rijn-Kennemerland (WRK nu Waternet/PWN)
Anno 2012 ca. 100 medewerkers verdeeld over de afdelingen:

4. • 2008-2009: Begin van een verschuiving binnen ons werkpakket van GC-MS
naar UPLC-MS voor polaire verbindingen zoals nieuwe bestrijdingsmiddelen,
hormonen, geneesmiddelen en daarnaast ook het overzetten van oude
methodes zoals chloorfenoxycarbonzuren, glymes, halogeenazijnzuren etc.
• Het project is in 2010 gestart met een evaluatie van verschillende Triple Quads
van 3 verschillende leveranciers op basis van door ons geleverde standaarden
en drink- en oppervlaktewaters met representatieve addities.
• Na evalutie bleek dat de TSQ voor HWL de beste keuze was vanwege de hoge
gevoeligheid (gem. 50x lager dan de concurrerende systemen).
• De keuze is definitief gemaakt door een FAT en SAT uit te voeren maar nu met
een factor 10 lagere addities.
• Installatie TSQ eind april 2011

5. Methodeontwikkeling – wat wil HWL bereiken?
• Methodeontwikkeling voor NP herbiciden, Trifosfaten, Chloorbenzenen, OCB/PCB
en de PAK (!)
• Planning oplevering van gevalideerde methoden juni 2012
(rekening gehouden met vertraging voor het leren kennen van de apparatuur,
software, vakanties, tegenslagen, optimisme van het leidinggevenden c.q. MT)
• Universele voorbewerking m.b.v. automatische SPE
(extra ontwikkeling Gilson SPE apparatuur)
• Uitgaan van de oplosmiddelen voor de huidige monsteropwerking:
- NP/trifosfaten: diethylether
- OCB/PCB: pentaan
- PAK acetonitril pentaan
• Monstername: door de lage detectiegrenzen is in combinatie met groot-volume PTV
injectie minder monster nodig.
6. Methodeontwikkeling
• Universele voorbewerking m.b.v. automatische SPE
- Alles met diethylether elueren: werkt niet voor alle componenten
- Alles met pentaan elueren: idem
- Eerst elueren met diethylether, daarna met pentaan
werkt …… maar ….
Voor de automatische SPE: eerst alle (‘vuile’) monsters filtreren
i.v.m. verstopping van de pompen maar dit werkt niet voor de
apolaire verbindingen zoals PAK’s …
7. Uiteindelijke methoden
• NP en trifosfaten m.b.v. SPE
100 ml monster, elutie met diethyleter, 50 µl PTV injectie
• Chloorbenzenen/OCB/PCB/PAK
50 ml in vial extractie met pentaan, 50 µl PTV injectie
11. - Door het ontbreken van een chromatogram en massaspectra veel minder zicht
op de injectie en de chromatografie, m.n. de conditie van de kolom.
- Nog geen historie van het instrument. Intern het lab geen vergelijking mogelijk
met een zelfde soort instrument.
Oplossing: TSQ historie opbouwen in full scan (Xcalibur) m.b.v. een
performance standaard waarvan de historie op een andere GCMS i.c.m. GV-PTV
al bekend is.
De TSQ 2-3 per week tunen, kalibreren en een performance injectie doen.
Specifieke massa’s voor componenten en oplosmiddel in 1 chromatogram
uitzetten
14. 2e
hindernis – keuze overgangen
• Geleverde bibliotheek met overgangen is nooit compleet voor jouw analyse
• Laat het denken in massaspectra los: een overgang van massa 200 naar 150
van een molecuul niet wil zeggen dat een eveneens voorkomende massa 202
binnen hetzelfde molecuul een overgang naar 152 heeft
• Lage gevoeligheid ondanks de bibliotheek? Zelf overgangen bepalen.
• Nieuwe overgangen gemaakt met standaarden 0,2 µg/l, eis voor elke overgang
voor kwantificering ca. 5*e5 counts
• Neem niet teveel nieuwe componenten tegelijk voor nieuwe overgangen (5-6)
• Overgangen bepalen kost tijd, neem er in het begin ook de tijd voor, speel
ermee, hou er rekening mee dat overgangen van een nieuwe verbinding een
andere kan verstoren (triazines)
• Hoeveel overgangen per component?

• ISO norm in ontwikkeling voor de identificering van doelstofcomponenten m.b.v.
chromatografie en massaspectrometrie dus voor UPLC-MS en GCMS

• Eisen Waterleidingbesluit:

- Als additie voor het bepalen van de AG wordt 0,02 µg/l geaddeerd.
• Probleem:
- De vorige versie van het Waterleidingbesluit geeft aan dat de AG wordt bepaald
op herhaalbaarheidscondities

- De nieuwe versie van het Waterleidingbesluit (de drinkwaterregeling) verwijst naar
de NEN 7777 (2011) welke aangeeft dat de AG wordt bepaald op

41. Validatieresultaat
• Tijdens de validatie is gebleken dat voor elke component een lage a.g. kan
worden gehaald door het hoge oppervlak van de pieken van de overgangen en de
S/N hiervan.
• Chloorbenzenen/OCB/PCB/PAK gevalideerd:
Juistheid en precisie voldoen aan de gestelde eisen in het Waterleidingbesluit
De gemeten AG’s zijn laag maar door een maximale reproduceerbaarheid van
8 verschillende series door 3 analisten gemaakt en gemeten op 8 verschillende
dagen met verschillende tuningen en ion volumes is de spreiding groter geworden
ORG is gestegen!
• Dit probleem wordt voorgelegd aan de Inspectie Drinkwaterbedrijven, intussen
heeft HWL alsnog deze ORG bepaald op herhaalbaarheidscondities, deze zijn
inderdaad beter!

43. Wat betekent het uiteindelijk in de praktijk?
• Van 3 naar 1 …
- Het is gelukt om een aantal analyses van GCMS singlequad , GCECD en HPLC
over te brengen op de TSQ.
Voordeel 1: De investering in een TSQ < GCMS+GCECD+HPLC.
Voordeel 2: Voor de nieuwe voorbewerkingsmethodes geldt dat er veel minder monster
nodig is. I.p.v 3100ml nu naar een vial van 66ml en een flesje van 100ml.
Dus lagere kosten,(disposable) glaswerk, eenvoudigere monstername.
Voordeel 3: (Chloorbenzenen, PAK, OCB/PCB): 50x minder extractievolume van
oplosmiddel, dus lagere kosten en minder belasting voor het milieu. De
voorbewerking is sterk vereenvoudigd en ca. 10x zo snel geworden.
De analysetijd is meer dan gehalveerd en teruugebracht naar 30 minuten.
Nadeel: Kwetsbaarheid, apparaat plat, 4 analyses plat … oplossing zoeken in
servicecontract of …?
44. Tenslotte …
• Bouw een historie op van je instrument, dit zegt veel over de staat van het
systeem omdat je de chromatografie in full scan mist. Is er eenmaal een historie,
hou deze regelmatig bij.
• Neem de tijd om het apparaat in de vingers te krijgen, let op stabiliteit van de
ionvolumes, doe af en toe een handmatige optimalisatie ter controle.
• Ervaring is dat MS-MS inderdaad simpel is, de TSQ is een werkpaard …

Produktie van ca. 120000 analyseresultaten per jaar
Voornaamste afdelingen: Chemie (organisch/anorganisch/metalen), Biologie (microbiologie en hydrobiologie), Advies (o.a. ontzorging legionella) en Logistiek (planning en monstername)
Een van de resultaten van deze fusie was dat er een grote hoeveelheid apparatuur ter beschikking kwam waardoor de eerste jaren weinig tot geen nieuwe investeringen werden gedaan
De laatste jaren is er een verschuiving begonnen in de vraag om meer polairdere stoffen te analyseren met als gevolg dat er in 2008 een 1e UPLC-MS werd aangeschaft en in 2009 een 2e. Tevens was het idee om GC-MS analyses zoals chloorfenoxycarbozuren en halogeenazijnzuren naar de UPLC MS over te brengen en mogelijk ook andere analyses.
Maar gezien de ontwikkelingen en ervaringen met UPLC-MS blijver er ook nog doelstofanalyses op de GCMS nodig.
