BACTERIAL DISPLAY TECHNOLOGY
Multiple cell surface display
platforms on bacteria have
been established. For display
on Gram-negative bacteria,
insertion scaffolds (based on
OmpA, OmpX, FhuA, FliTrx,
FimH), N-terminal display
scaffolds (based on EstA,
AIDA-I, IgA protease), C-ter-
minal display scaffolds (based
on Lpp-OmpA, Invasin), and
combination of N-terminal
and C-terminal display using
circularly permuted OmpX
(CPX) could be chosen. For
Gram-positive bacteria,
foreign proteins could be
displayed via covalently
anchored proteins including
SPA and FnBTB.
The protein or peptide library
is constructed using recombi-
nant DNA technology and
displayed on bacteria. The
bacterial library can be
labeled with fluorescently
labeled target protein
(antigen), or incubated with
biotin-tagged target protein
and streptavidin-magnetic
beads. Target-binding bacte-
ria are isolated using fluores-
cence-activated cell sorting
(FACS) or magnetic-activated
cell sorting (MACS). The
selection cycle is generally
repeated, and selected bacte-
ria are typically amplified by
growth in between the cycles.
Finally, isolated binders are
screened using whole-cell
ELISA-based methods, identi-
fied using DNA sequencing
and subcloned for soluble
production and detailed
characterization.
