THE PROBLEM WITH SALMONELLA & LISTERIA
IN FOOD PRODUCTS
Hence, a variety of methods have been developed for rapid detection of foodborne
pathogens as it is required in many food analyses. Most of them require an enrichment step
before being able to apply the detection device.
For all these reasons, FoodChek has developed Actero™ Salmonella Enrichment Media and
Actero™ Listeria Enrichment Media to selectively enrich Salmonella and Listeria in a variety

Salmonella spp. (species) is the leading cause of food infection in the United States, with an
estimated number of Salmonella infections of 1.4 million cases per year, and causes the
second largest number of human food-borne illnesses in Europe. Listeriosis caused by
Listeria monocytogenes (L. mono) is low in incidence but its mortality rate is high placing
the microorganism to the top of pathogens contributing to food-borne illness and resulting
in death. Older adults, pregnant women and their fetuses, newborn infants, and other
persons with reduced immune function are particularly at risk for invasive listeriosis.
Salmonella infections have been associated with eating foods, such as meat, eggs and fresh
produce contaminated with animal or human feces. The main causes of Salmonella illness
are poultry and eggs. Listeria monocytogenes has the potential to be present in all raw
foods. Cooked foods can also be contaminated and is usually as the result of post-process
contamination. The pathogen has been isolated from a very wide range of processed foods
includingincluding pâtés, milk, soft cheeses, ice cream, ready-to-eat cooked and fermented meats, smoked and lightly processed fish products and
other seafood.

Abstract
FoodChek Systems Inc., in collaboration with a Real-Time PCR Assay manufacturer, has developed effective methods for easy and rapid
detection of Salmonella and Listeria in a variety of environmental and food samples. The methods has been successfully granted AOAC
PTM CERTIFICATION. The results of the study demonstrated that performance of the candidate methods is statistically equivalent to the
reference culture method for detecting Salmonella or Listeria in food. The time to detect Salmonella spp. has been reduced up to several
hours, depending the matrix, including the enrichment and detection steps when the Actero™ Salmonella Enrichment Media was used.
Similar observation was obtained for the detection of Listeria in food using Actero™ Listeria Enrichment Media. No false negative or false
positive results were observed. The high accuracy and reliability of the methods were confirmed in the independent laboratory
validation study.

Results
Probability of detection (POD) statistical model was used to evaluate the differences between presumptive and confirmed results as well
as between the alternative and the reference methods.
The results of the different method comparison validation studies for variety of food products are summarized in the table below. POD
analysis didn’t show any significant differences (the 95% confidence interval of the dPODs contains zero) in the method performance
between the Real-Time PCR Assay and the reference methods.
Conclusions
The results of the study demonstrated that performance of the Real-Time PCR Assay method to detect Salmonella in a variety of food
samples enriched with Actero™ Salmonella Enrichment Media is equivalent to the reference method. It has been proven that Actero™
Salmonella Enrichment Media is able to selectively enrich Salmonella for its further detection in the shortest possible time.

Results

The methods has been successfully granted AOAC PTM CERTIFICATION. The results of the study demonstrated that performance of the candidate methods is statistically equivalent to the reference culture method for detecting Salmonella or Listeria in food. The time to detect Salmonella spp. has been reduced up to several hours, depending the matrix, including the enrichment and detection steps when the Actero™ Salmonella Enrichment Media was used.
