Wnt is regulated by endogenous antagonists among them Sclerostin which is a protein mainly produced by osteocytes. Many studies show an effect of bone remodeling on cartilage homeostasis. Therefore, in the present work, we assess the role of Sclerostin in cartilage maintenance and in OA.
Inhibition of the Wnt/B-catenin pathway by Sclerostin abolished the metalloproteinases increase induced by wnt and had a slight effect on the differentiation gene regulation. And finally Sclerostin was able to inhibit the hypertrophic phenotype induced by Wnt.
