This is a pretty exciting post to write as our Thermo Scientific EASY-nLC 1000 split-free, nano-flow UHPLC
system was awarded the Select Science Scientists' Choice Award for Best New Separations Product of 2011 at
Pittcon this year! You may not know but this is a reader-voted award, meaning that an overwhelming number of
Select Science readers found this to be an outstanding instrument! We are, of course, extremely pleased about this
award!
Here we present a case study showing how a customer increased protein and peptide identifications by almost
30% using this award-winning system.
Customer
The illustrious Center for Experimental Bioinformatics (CEBI), part of the Department of Department of
Biochemistry and Molecular Biology at the University of Southern Denmark (Odense, Denmark) is recognized
globally for its extensive contributions to the advancement of proteomics and focuses its research on fundamental
processes that control cell life and development, including how genes are structured and organized, how gene
activity is regulated and how gene information via ribonucleic acid (RNA) is translated into protein.
Challenges Faced
The CEBI was using conventional high performance liquid chromatography (HPLC) for its proteomics analyses
for which they used 75 µm i.d., 20 cm long columns. They wished to switch to ultra high performance liquid
chromatography (UHPLC) and thinner and longer columns for the benefits of improved separation and ionization
with no additional sample loss. But, there were two challenges:
• Using a high pressure technology could result in increased internal connection leaks.
• Manufacturing small i.d. columns can be a challenge.
Solution
In December 2010, we offered the CEBI team the opportunity to test the Thermo Scientific EASY-nLC 1000
split-free, nano-flow UHPLC system before the instrument’s official launch in June 2011. They took us up on the
offer and the rest is history as they say!
The instrument was able to seamlessly integrate with the two mass spectrometry systems at the Center and was
operational instantaneously! As per Lasse Falkenby, Research Assistant at CEBI, “Compared with other solutions
available on the market the EASY-nLC 1000 can use shorter gradients on 50 µm UHPLC columns with 1.9 µm
beads due to its market leading operating pressure, thereby delivering superior separation capability with razor-
sharp peaks."
Benefits
• The higher operating pressure range of our instrument resulted in about 30% more peptide and protein
identifications using longer columns and smaller beads.
• The system significantly shortened analytical cycles and increased productivity by accelerating sample
loading, washing and column re-equilibrations.
• The enhanced sensitivity of the small i.d. columns allowed the CEBI team to scale down experiments and
reduce costs.
