infarction
Acute myocardial infarction is
myocardial necrosis resulting from
acute obstruction of a coronary artery.
Symptoms include chest discomfort with
or without dyspnea, nausea, and/or
diaphoresis. Diagnosis is by
electrocardiography (ECG) and the
presence or absence of serologic
markers.
3. ROLE OF GDF5 IN CARDIAC RUPTURE.
GDF5(growth differentiation factor 5), helps prevent heart
rupture after a heart attack. In mice without GDF5, the risk of
heart rupture was lower, and survival rates improved. GDF5-
deficient mice had less heart weight relative to body weight,
increased scarring, and lower blood pressures, indicating reduced
stress on the heart. Despite changes in immune markers, bone
marrow transplantation suggested GDF5 from heart tissue, not
bone marrow, was vital in preventing heart rupture post-heart
attack. Overall, while GDF5 loss reduces heart rupture risk, it leads
to increased scarring and long-term negative heart changes.
4. The article aims to investigate the role of growth
differentiation factor 5 (GDF5) in preventing cardiac rupture
following acute myocardial infarction (MI) in mice. It builds on
previous research showing GDF5's impact on limiting infarct
expansion post-MI. The study explores the acute post-MI
effects of GDF5 deficiency and provides insights into reduced
cardiac rupture incidence, improved survival rates, and
associated physiological and molecular changes in GDF5-
deficient mice compared to wild-type mice. The findings
contribute to understanding the molecular factors involved in

METODOS
Se fijaron corazones en paraformaldehído al 4%, se procesaron en parafina y se
obtuvieron secciones transversales de 5 µm. Estas se tiñeron con hematoxilina y
eosina o tricrómicamente con azul de Masson y se visualizaron en un
microscopio AxioImager 2 (Zeiss, Alemania). Se midió el área transversal de los
cardiomiocitos utilizando ImageJ, promediando los resultados de nueve campos
de visión de alta potencia tomados de tres secciones cardíacas por ratón.
En el tercer día después del infarto de miocardio, se sacrificaron ratones y se perfundieron con
solución salina antes de extraer los corazones. Se microdisecó el ventrículo izquierdo en regiones
de infarto y no infarto, congelándolas rápidamente. Se aisló el ARN tisular con TRIzol, y se
sintetizó cDNA usando qScript cDNA SuperMix. Luego, se llevó a cabo qRT-PCR con igual
cantidad de cDNA para genes de interés y el gen de referencia, hipoxantina-guanina fosforribosil
transferasa, utilizando SYBR Green Master Mix en un LightCycler 480 de Roche.

6. METODOS
Tres días después del infarto de miocardio, los ratones fueron sacrificados y perfundidos con 10
mL de solución salina tamponada con fosfato (PBS) antes de la extracción del corazón. Se
microdisecó el ventrículo izquierdo en regiones de infarto y no infarto, y se congeló
rápidamente. Se prepararon lisados tisulares a partir de las regiones de infarto aisladas y se
corrieron en geles de poliacrilamida desnaturalizantes. Se realizaron Western blots utilizando
anticuerpos para TNF-α, Pro-COL1A1 y GAPDH (proteína de referencia).
Tres días después del infarto de miocardio, se recogió
sangre mediante punción cardíaca en microvetas de EDTA
de 100 µL. Las muestras fueron analizadas en un
hematómetro Hemavet (modelo 950FS, Drew Scientific) en
el Centro de Recursos Animales de la Red de Salud
