Introduction
• Clotting time is the time taken from puncture of
blood vessel to formation of fibrin thread.
• Clotting time determines the function of clotting
factors

AIMS
• To determine CT of blood by capillary glass tube
method
APPARATUS REQUIRED
• CAPILLARY GLASS TUBE
• SPIRIT
• COTTON
• LANCET
• STOP WATCH
Apparatus Required for CT
PROCEDURE
• Under aspetic precaution ,prick the finger
• Draw the blood immediately in to one end of capillary tube
. Blood flows into the tube by capillary action.
• When the tube is almost full, keep it between the palms for
2 minutes to maintain it at body temperature.
• Then break 1 cm of the tube . Make successive breaks
every 30 seconds
OBSERVATION
• Look for a Fibrin thread between the broken ends,
which indicates CT
Normal CT by capillary glass tube method is 3 – 8
Minutes.
Result
• Clotting time by capillary glass tube
method ____________ Minutes.
CT Prolonged
• Haemophilia A,B,C
• Vitamin K deficiency
• Livers disorders
• Overdose of anticoagulants like warfarin or Heparin.
Determination of Blood Group
Aim
•To determine my blood group
APPARATUS REQUIRED
•Porcelain tile
•Anti sera A,B and D
•Lancet
•Spirit
•Cotton
•Toothe sticks
Apparatus Required for Blood grouping
PRINCIPLE
The surface of the red cell membrane contains
agglutinins( antibodies) . The red cells are allowed
to react with commercially made antibodies . The
presence/absence of agglutination (clumping of
RBCs) determines the blood groups.
PROCEDURE
• Divide the porcelain tile into 4 portions (A,B,C & D)
• Place blood drop into 4 portions in the tiles
• Then place a drop of Anti sera A, B and D in three
portions ( A,B and D)
• Forth corner C is the control cell suspension used for
comparison.
• Mix thoroughly with separate sticks and gently rock
the tile
OBSERVATION AND RESULT
• Wait for 8 – 10 minutes
• Look for agglutination ( this is the time required for
antigen-antibody reaction)
