This document discusses an algorithm for image registration of physical slices with discontinuities in microscopy. Standard registration algorithms assume a continuous deformation field, which does not account for discontinuities between slices. The authors propose minimizing an L1-total variation functional using a piecewise monotonic subgradient algorithm. This convex optimization approach iteratively minimizes the dual problem and achieves better registration results than graph cut-based methods, with lower error and visually acceptable results even where graph cut fails. Experimental results show the subgradient method consistently outperforms graph cut.
