ChromaDex Provides Update on $62 Million Ingredient Agreement with Network Marketing
Company 5LINX®

four-year ingredient supply and brand licensing agreement with 5LINX®, one of the largest and

5LINX® plans on launching several new innovative health and wellness products that feature
ChromaDex's recently launched patented NIAGEN™ nicotinamide riboside (NR) and patented
PURENERGY® ingredients at its upcoming international event in Atlantic City, NJ. The event
will be held May 14 to 17 and is expected to be attended by thousands of 5LINX® sales

In January 2014, 5LINX® was granted marketing rights for ChromaDex's recently launched
patented NIAGEN™ nicotinamide riboside (NR) for use in dietary supplements exclusively in
the network marketing sales channel in the United States, Canada and the Philippines. Under the
terms of the agreement, 5LINX® must purchase from ChromaDex $2.1 million of NIAGEN™ in
2014, and an aggregate of $46 million of NIAGEN™ from 2015 through 2017. NIAGEN™ is
the first and only commercially available brand of NR, which is found naturally in trace amounts
in milk and other foods and is a more potent, no-flush version of niacin (vitamin B3). Published
research has shown that NR is perhaps the most potent precursor to NAD+ in the mitochondria
of animals. NAD+ is arguably the most important cellular co-factor for improvement of
mitochondrial performance and energy metabolism.
5LINX® was also granted marketing rights for ChromaDex's patented PURENERGY® ingredient
for use in dietary supplements, with select exclusivity in the network marketing sales channel in

About Nicotinamide Riboside (NR):
Sometimes referred to as the "Miracle Molecule" or "Hidden Vitamin," NR is found naturally in

Published research has shown that NR is perhaps the most effective NAD+ booster, an essential

A study by researchers from Harvard Medical School in conjunction with the National Institute
on Aging was published in December 2013 in CELL demonstrated that mitochondrial
dysfunction (a hallmark of aging) in aging mice is due to a disruption in Sirtuin1-dependent
nuclear-mitochondrial communication. They further show that a reduction in NAD+ levels is
responsible for this disruption. Excitingly, they also show that this mitochondrial dysfunction is
readily reversible by administration of a NAD+ precursor. The authors show that "1 week of
treatment with a compound that boosts NAD+ levels is sufficient to restore the mitochondrial
homeostasis and key biochemical markers of muscle health in a 22-month-old mouse to levels
similar to a 6-month-old mouse."

It's time for my Thursday update.
Once again, I've taken 15 Niagen capsules so far today, with another 5 reserved for bedtime. (I
didn't mention the fact that I take two 100-mg. capsules of the new Xymogen bioabsorbable s-
acetyl glutathione capsules every day along with a transdermal glutathione cream, and 8-hour,
sustained release n-acetyl cysteine capsule at bedtime.)
Once again, I headed for the Wellness Center this morning. But it has occurred to me that I've
probably added a few pounds over the past two months. I've been concerned about getting
enough protein to support the rebuilding of muscles, but that mindset may have invited extra
calories. (I don't weigh myself very often, since I'm not normally focused on losing weight.)
Chinning myself is a sensitive function of how heavy I am. A couple of pounds can make a
sizable difference. And since we're trying to determine whether Niagen is making meaningful
differences, my weight needs to be tightly controlled. I'll check my weight when I wake up
tomorrow morning, and try to lean toward a weight-loss mode over the next few weeks.
At the Wellness Center this morning, I tried for the first time elevating the treadmill. I found that
at a slope of 10°, I was generating 7.4 METs at 3 miles an hour, and 8 METs at 3.3 miles an
hour. I ran about ½ mile at 3 mph. (7,4 METs) and about ½ mile at 3 mph (8 METs). My pulse
rate gradually climbed from the 90's to the low 120's. I ran another mile, and then began running
60-second sprints at 9,5 METs, resting about two minutes between sprints... except that today, I
raised the bar a bit by running 120-second sprints. My highest pulse rate was 130, which is
below my maximum pulse rate by any standards. (I've been reticent about pushing myself harder
without anyone watching my EKG.) I was breathing hard enough to make it hard to carry on a
conversation by the time I finished.
Last night, I began investigating to find out what the norms are for someone my age. Here they
are:
Heart Rate:
I found two formulas for this. The classical formula is 220 - (your age). For me, at 85, that
works out to be 135 beats per minute.
Another formula that purports to update the above formula uses 208- 0.7(your age). This
formula yields 148.5 beats per minute.
Maximum Level of Exertion (METs Level):
For someone who's athletically conditioned, the maximum treadmill METs level is given by
18.7- 0.15 X your age. For me at 85, that works out to be 5.95 METs. Of course, I ran today for 6
minutes at 8 METs without reaching my maximum heart rate.
If I use 8 METs as my maximum level of exertion, subtract 5.95 METs from it, and divide by
0.15 to arrive at an age-equivalent METs level, I arrive at 2.05/0.15 = 13⅔ years. Subtracting this
from 85 produces an equivalent "physiological age" of 71⅓ years.
Of course, I ran two minutes at 9.5 METs without reaching my maximum heart rate. If I used 9
METs (conservatively( as my peak treadmill level of exertion, I would arrive at an offset of
3.05.0.15 = 20⅓ years. Subtracting this from 85 gives a physiological age of 64⅔ years.
Of course, this doesn't quite give the picture. Our 7½-year-old darling and I run races and play
tag and Frisbee and hide-and-go-seek (including running for home base). Our 40-year-old
daughter, Lisa, died in her sleep in December, 2006, and left her three-month-old baby, Amber,
for my wife and I to rear, so we've been back on the "Daddy and Mommy" track.
After running on the treadmill, I went to the "Chin-Lift" and tried chinning myself. I was just
barely able to pull myself up with 4 pounds of lift. At 10 pounds of lift, I handily chinned myself
12 times. quitting before I quite reached my limit.
My athletic fitness testing in January when I first approached the Wellness Center put me in the
95th percentile for VO2 max at about 30 (Mkm). I inquired about the age group to which that
percentile ranking applied, but the personal trainer who administered the January fitness exam.
didn't know. On the Partial Curl-Up (Rep) ranking, I was in the 99th percentile in January. I was
also in the 99th percentile on the waist-to-hip ratio. I was in the 89th percentile for body fat
composition. On other measures testing flexibility, etc., I didn't fare as well.
Presumably, my workouts at the Wellness Center will improve some of these benchmarks over
time.
The verdict for Niagen? It depends upon how long it takes Niagen to do its thing in humans, and
it depends upon how much weight (if any) I may have added since the last time I weighed
myself. There hasn't been any "Wow!" boost in my performance over the past 48 hours.
There is a lot of buzz recently in anti aging forums surrounding a new study from David
Sinclair of resveratrol fame

Niacinimide Riboside ( Niagen ) has been shown to come close to what Sinclair used to
boost NAD, more so than regular niacin. There is evidence that CD38 reduces NAD+ and
inhibiting it with flavonoids like C3G, apigenin etc is synergistic.

I think that a supplement combining the two would be important for LEF to consider. I am
already supplementing this on my own with what is available. The problem with flavonoids
for CD38 inhibition is bioavailability ....this is where LEF's expertise could come into play

The use of the flavonoids is interesting for the support of NAD. The use of Niagen is over
rated. I bought a couple of bottles of this product early on and have since discovered that it
is only twice as potent as niacinamide as far as increased NAD. The Niagen is expensive and
the capsules are only 100 mg in their max dosage cap. However, nacinamide is much more
effective than niacin in obtaining higher NAD levels.
The discovery itself is a further update and some clarification of old news for the use of
niacinamide with resveratrol. Go to Life Enhancements site type in Krebs cycle or
niacinamide and see that they have been recommending the use of niacinamide since the
early MIT studies years ago.

Somebody correct me if I'm wrong here, but isn't there some independent benefit of the
"niacin flush" you can only get from niacin?
I long ago read some health guru make the pitch about how the niacin flush will clean your
arteries and otherwise does all sorts of amazing things for your cardiovascular system that
cannot come from niacinamide.
I make a point to subjecting myself to an intense, red, all body flush with a treatment of
about 750 mg of quick-release niacin taken once per month (in addition to daily, regular
supplementation).
Anyone care to advise me further/otherwise?

Sounds good but its a three things you should be taking maybe four with niacin
Q10 blend with Omega-33333
Resveratrol
DHEA
Good multi -calcium good amount of Vit D

Nicotinamide riboside is different from Niacinimide or niacin, and is a more direct NAD+
activator. It's mechanism of action has nothing to do with the flushing effect that niacin
users want to increase HDL.
It's the closest one can get to the molecule used in the Sinclair study.
" In eukaryotes, nicotinamide riboside is a newly discovered NAD+ precursor that is
converted to nicotinamide mononucleotide by specific nicotinamide riboside kinases, Nrk1
and Nrk2. In this study, we discovered that exogenous nicotinamide riboside promotes Sir2-
dependent repression of recombination, improves gene silencing, and extends lifespan
without calorie restriction. "

Just so readers don't get confused nicotinamide and niacinamide are two different terms
used interchangeably to describe the same thing. Nicotinamde riboside is a form of B3 found
in milk and beer that is effective in increasing NAD. Niagen is the commercially available
product for nicotinamide riboside.

Interesting read. The author is familiar , I think he also writes for LEF. The Leonard
Guarante lecture should be interesting. David Sinclair did his doctoral work under his
supervision I believe.

In the buzz online about Niacinimide Riboside, a lot of mentions of it occurring naturally in
beer and milk. Obviously, nobody is going to experience any of the experimental-level type
of mouse-rejuvenating effects from doses naturally found in food.
But just as a curiosity, does anybody have data on exactly how much is contained in these
foods?
Beer appears to contain about 1.8 mg of niacin per serving, as does a couple of tablespoons
of brewer's yeast. Now, is the naturally-occurring niacin therein all or mostly in this unique
form? Brewers yeast is one of those healthy health foods long touted since the beginnings
of the "good food" movement of the late 1920's., I'm wondering about the niacin in it as
one of its many good-for-you factors that people since long ago have made testimonials
about?
Personally, I find brewer's yeast a bit repulsive. Last time I had some on hand, I ended-up
feeding my compost pile the bulk of it and had decided that would be the last time I would
ever try that nasty stuff again, but now I'm giving it another look.

the study did.

Interesting. I have read somewhere that NAD itself is too large of a molecule to be
absorbed. Perhaps their delivery system gets around that. There isn't any reliable data
regarding NAD enhancement in humans with any specific product or dosage of
products. According to Guerante and Sinclair, NR being a small molecule is the best and
most direct precursor of NAD
This is another reason that it would be interesting to see how LEF addresses this, and which
molecule they will choose to use. The problem with anything other than plain old niacin, is
patent issues.

Niagen is NR cholride and not NR. I suspec the chloride form has bioactive differences than
NR. NR is very expensive. NR chloride is much less expensive.

In light of this information on the importance of NAD, would anyone advise if one should
continue taking an NADH supplement? It sounds like this pseudohypoxia cell-aging occurs
with imbalanced NADH at the mitochondrial cell level, so is taking a dietary supplement of
NADH therefore somewhat counterproductive to the goals of life extension?

interesting is the fact that NAD ratio's cannot yet bemeasured but if one reads the beginning of
the document there is mention thatas one ages there is possible degradation in the pathway so it
might be aproblem for many and supplementation may have some benefit or unknown affect.
now on page 21 cited above there is "This balance NAD+:NADH maybe moreabout...."

I hope LE follows up with more data regarding the bioavailability and pharmakinetics of NR
and any studies that show NR taken orally increases NAD+ in humans, and if so, at what
dose. Anecdotally most people on various blog sites feel that somewhere between 500-
1000 mg's is where they feel something. I take 625 mg, but am awaiting actual real
data......

Yes, this is what I was referring to. Probably, as with many supplements, you can "feel"
something if you are in a status of nutrient deficiency and/or have a pathology while, when
this is not the case, you put trust in the studies which show benefit and you balance with
risks. That is also why it is important to find (healthy) human studies. For NR do I
understand correctly ChromaDex is also coming up with a optimal human dose? Of course I
would like to read LEF on this. It reminds me a bit the discussions on trans-resveratrol
dosage when people also mega-dosed while you also had studies showing benefits at
relatively low doses.
The key is to establish whether or not taking NR orally actually raises NAD, and how much is
necessary. The studies that Chromadex is currently conducting are looking at safety and
pharmakinetics, which are different.
It seems like that's all pointless if orally administered NR doesn't raise NAD significantly.
I guess we hope, and wait.....

I'm also really interested in the optimal dosage of NR, and best means of restoring NAD+
levels. I'm hoping that the Chromadex studies will reveal this soon, because it certainly
would be a lot of wasted money if NR isn't orally bioavailable.
Anyway, i've been posting around a bit because I recently finished a month-long trial of
Biotivia's MitoTrans, which is their stabilized NAD+ supplement. Ive tried both niagen and
MitoTrans, in attempts of improving my chronic fatigue, lack of motivation, and low physical
endurance.
I can't say I really noticed all that much from niagen - very slight improvement in energy
maybe, and no change in body composition or really any other noticeable effects.
With MitoTrans, while the effects are subtle, i seem to have lost a pretty significant amount
of body fat and have a noticeably improved capacity for endurance exercise (like walking for
long distances or jogging). I admit that these could possibly be placebo, but I am very
impressed by the reduction in body-fat that ive noticed. i wish i had taken some before and
after pictures, but i'm not sure how useful that would have been since i've also started a
consistence weightlifting routine.
I would be very interested in knowing how MitoTrans compares to Niagen in terms of
bioavailability and raising of NAD+ levels. From what i understand, when NR is ingested as
precursor of NAD+, it is always NMNAT enzyme dependent pathway. and direct
supplementation of NAD+ is not enzyme dependent. so I would assume that MitoTrans is a
good bit more effective than niagen. At least, this was my rationale for trying it.

I started to take Niagen this morning. I'm curious if I'll see any results as I'm 45. Some
people younger than me said that they had "visible" results with Niagen. I hope that is the
case as I do not like this to be another overhyped supplement.
Besides Biotivia (which is quite expensive $475 for one month!), there is RevGenetics that
will have a new product on the market.
I wonder if LEF is conducting any medical research in this area of NAD+?
The way I see it, is that once some rejuvenation pathway is identified, it is worth exploring
it and improve upon initial discoveries, either by delivery mechanism, or new molecules,
etc. And LEF do have the resources to take on such tasks. Similar LEF can look into GDF-
11, MitoQ , SkQ1 or TA-65.
I wonder why LEF is not conducting any research with these factors/molecules?

purveyors. I basically added it for the anti-aging science, but noticed something
positive. Normally, at the end of the day after work I tend to feel inclined to either take a
short nap, or lay on the sofa and watch TV for bit just to relax and decompress after the
stress of the day. I noticed almost a day or two after adding the NAD formula to my
regimen that I would continue on without the desire to lay down or take a nap. There was
nothing new other than that formula that changed in my diet, vitamins, routine, etc., so I
new it had to be the NAD formula. I have been on it for weeks and I still have that constant
energy state, not a revved up state, but just a smooth lasting energy. Has any one else
experienced this using the Niagen?

I am trying some from live cell research, it's 250 mg of Niagen. Right now I'm into my
second day.
The first day I felt a little tired, more than usual. The second day, the same.
I will keep monitoring the results, two days isn't enough and could be coincidental.

i've been looking over the internet for users reviews of Niagen. At first there were some that
felt results.
Some of the users went up to a 1000 mg dose.
Now after awhile there is hardly any one reporting any kind of noticeable benefit, including
this forum.
I am taking 250 mg first thing in the morning, and now have bumped it up to 500 mg.
Still not noticing benefit but maybe it is doing some good, don't know.
Has anyone received noticeable benefit as yet?

impact lipids, despite a recent study that concluded that niacin had no protective effect (and
it may not have one in some people). The histamine-releasing effect of niacin has other
benefits. Niacinamide and niacin also may have longevity benefits, yet niacinamide's effect
in this area has been argued both ways.

Once again, I've taken 15 Niagen capsules so far today, with another 5 reserved for bedtime. (I didn't mention the fact that I take two 100-mg. capsules of the new Xymogen bioabsorbable s- acetyl glutathione capsules every day along with a transdermal glutathione cream, and 8-hour, sustained release n-acetyl cysteine capsule at bedtime.) Once again, I headed for the Wellness Center this morning. But it has occurred to me that I've probably added a few pounds over the past two months. I've been concerned about getting enough protein to support the rebuilding of muscles, but that mindset may have invited extra calories. (I don't weigh myself very often, since I'm not normally focused on losing weight.) Chinning myself is a sensitive function of how heavy I am. A couple of pounds can make a sizable difference. And since we're trying to determine whether Niagen is making meaningful differences, my weight needs to be tightly controlled. I'll check my weight when I wake up tomorrow morning, and try to lean toward a weight-loss mode over the next few weeks. At the Wellness Center this morning, I tried for the first time elevating the treadmill. I found that at a slope of 10°, I was generating 7.4 METs at 3 miles an hour, and 8 METs at 3.3 miles an hour. I ran about ½ mile at 3 mph. (7,4 METs) and about ½ mile at 3 mph (8 METs). My pulse rate gradually climbed from the 90's to the low 120's. I ran another mile, and then began running 60-second sprints at 9,5 METs, resting about two minutes between sprints... except that today, I
  • 6. raised the bar a bit by running 120-second sprints. My highest pulse rate was 130, which is below my maximum pulse rate by any standards. (I've been reticent about pushing myself harder without anyone watching my EKG.) I was breathing hard enough to make it hard to carry on a conversation by the time I finished. Last night, I began investigating to find out what the norms are for someone my age. Here they are: Heart Rate: I found two formulas for this. The classical formula is 220 - (your age). For me, at 85, that works out to be 135 beats per minute. Another formula that purports to update the above formula uses 208- 0.7(your age). This formula yields 148.5 beats per minute. Maximum Level of Exertion (METs Level): For someone who's athletically conditioned, the maximum treadmill METs level is given by 18.7- 0.15 X your age. For me at 85, that works out to be 5.95 METs. Of course, I ran today for 6 minutes at 8 METs without reaching my maximum heart rate. If I use 8 METs as my maximum level of exertion, subtract 5.95 METs from it, and divide by 0.15 to arrive at an age-equivalent METs level, I arrive at 2.05/0.15 = 13⅔ years. Subtracting this from 85 produces an equivalent "physiological age" of 71⅓ years. Of course, I ran two minutes at 9.5 METs without reaching my maximum heart rate. If I used 9 METs (conservatively( as my peak treadmill level of exertion, I would arrive at an offset of 3.05.0.15 = 20⅓ years. Subtracting this from 85 gives a physiological age of 64⅔ years. Of course, this doesn't quite give the picture. Our 7½-year-old darling and I run races and play tag and Frisbee and hide-and-go-seek (including running for home base). Our 40-year-old daughter, Lisa, died in her sleep in December, 2006, and left her three-month-old baby, Amber, for my wife and I to rear, so we've been back on the "Daddy and Mommy" track. After running on the treadmill, I went to the "Chin-Lift" and tried chinning myself. I was just barely able to pull myself up with 4 pounds of lift. At 10 pounds of lift, I handily chinned myself 12 times. quitting before I quite reached my limit. My athletic fitness testing in January when I first approached the Wellness Center put me in the 95th percentile for VO2 max at about 30 (Mkm). I inquired about the age group to which that percentile ranking applied, but the personal trainer who administered the January fitness exam. didn't know. On the Partial Curl-Up (Rep) ranking, I was in the 99th percentile in January. I was also in the 99th percentile on the waist-to-hip ratio. I was in the 89th percentile for body fat composition. On other measures testing flexibility, etc., I didn't fare as well. Presumably, my workouts at the Wellness Center will improve some of these benchmarks over time. The verdict for Niagen? It depends upon how long it takes Niagen to do its thing in humans, and it depends upon how much weight (if any) I may have added since the last time I weighed

The use of Niagen is over rated. I bought a couple of bottles of this product early on and have since discovered that it is only twice as potent as niacinamide as far as increased NAD. The Niagen is expensive and the capsules are only 100 mg in their max dosage cap. However, nacinamide is much more effective than niacin in obtaining higher NAD levels. The discovery itself is a further update and some clarification of old news for the use of niacinamide with resveratrol. Go to Life Enhancements site type in Krebs cycle or niacinamide and see that they have been recommending the use of niacinamide since the early MIT studies years ago. Obviously, nobody is going to experience any of the experimental-level type of mouse-rejuvenating effects from doses naturally found in food. But just as a curiosity, does anybody have data on exactly how much is contained in these foods? Beer appears to contain about 1.8 mg of niacin per serving, as does a couple of tablespoons of brewer's yeast. Now, is the naturally-occurring niacin therein all or mostly in this unique form? Brewers yeast is one of those healthy health foods long touted since the beginnings of the "good food" movement of the late 1920's., I'm wondering about the niacin in it as one of its many good-for-you factors that people since long ago have made testimonials about? Personally, I find brewer's yeast a bit repulsive. Last time I had some on hand, I ended-up feeding my compost pile the bulk of it and had decided that would be the last time I would ever try that nasty stuff again, but now I'm giving it another look. Adrian Interesting. I have read somewhere that NAD itself is too large of a molecule to be absorbed. Perhaps their delivery system gets around that. There isn't any reliable data regarding NAD enhancement in humans with any specific product or dosage of products. According to Guerante and Sinclair, NR being a small molecule is the best and most direct precursor of NAD This is another reason that it would be interesting to see how LEF addresses this, and which molecule they will choose to use. That is also why it is important to find (healthy) human studies. For NR do I understand correctly ChromaDex is also coming up with a optimal human dose? Of course I would like to read LEF on this. It reminds me a bit the discussions on trans-resveratrol dosage when people also mega-dosed while you also had studies showing benefits at relatively low doses. The key is to establish whether or not taking NR orally actually raises NAD, and how much is necessary. The studies that Chromadex is currently conducting are looking at safety and pharmakinetics, which are different.   • 14. I'm also really interested in the optimal dosage of NR, and best means of restoring NAD+ levels. I'm hoping that the Chromadex studies will reveal this soon, because it certainly would be a lot of wasted money if NR isn't orally bioavailable. Anyway, i've been posting around a bit because I recently finished a month-long trial of Biotivia's MitoTrans, which is their stabilized NAD+ supplement. Ive tried both niagen and MitoTrans, in attempts of improving my chronic fatigue, lack of motivation, and low physical endurance. I can't say I really noticed all that much from niagen - very slight improvement in energy maybe, and no change in body composition or really any other noticeable effects. With MitoTrans, while the effects are subtle, i seem to have lost a pretty significant amount of body fat and have a noticeably improved capacity for endurance exercise (like walking for long distances or jogging). I admit that these could possibly be placebo, but I am very impressed by the reduction in body-fat that ive noticed. i wish i had taken some before and after pictures, but i'm not sure how useful that would have been since i've also started a consistence weightlifting routine. I would be very interested in knowing how MitoTrans compares to Niagen in terms of bioavailability and raising of NAD+ levels. From what i understand, when NR is ingested as precursor of NAD+, it is always NMNAT enzyme dependent pathway. and direct supplementation of NAD+ is not enzyme dependent. so I would assume that MitoTrans is a good bit more effective than niagen. Some people younger than me said that they had "visible" results with Niagen. I hope that is the case as I do not like this to be another overhyped supplement. I wonder if LEF is conducting any medical research in this area of NAD+? The way I see it, is that once some rejuvenation pathway is identified, it is worth exploring it and improve upon initial discoveries, either by delivery mechanism, or new molecules, etc. And LEF do have the resources to take on such tasks. I am trying some from live cell research, it's 250 mg of Niagen. Right now I'm into my second day. The first day I felt a little tired, more than usual. The second day, the same. At first there were some that felt results. Some of the users went up to a 1000 mg dose. Now after awhile there is hardly any one reporting any kind of noticeable benefit, including this forum. I am taking 250 mg first thing in the morning, and now have bumped it up to 500 mg. Still not noticing benefit but maybe it is doing some good, don't know.
