Eddy’s Hot Plate analgesiometer

Aim
• To demonstrate the analgesic effect of Opiod analgesics
in rodents using Eddy’s hot plate analgesiometer.
Principle
• Rodents will show aversion effect to thermal
stimulus by paw licking & jumping after placing
onto the hot surface
• Source of pain: Contact heat
• Endpoint: Paw licking and jumping
• Cut off time : Mouse - 15 – 20 sec
Rat - 20 – 30 sec
Materials
• Weighing balance
• Eddy’s hot plate analgesiometer
• Rats
• Drug: Tramadol
• Needle: 24 gauge
• Gloves

• Indicator lights:
1) TEMPT: Indication of heating is on
Light blinking - heating process to achieve the
temperature is going on
Light stable - set temperature is achieved
2) LID: Indication of Lid of the plate is open
Keys -
• OK: To enter the set parameters
• :To move forward or to increase the value
• <: :To move backward or to decrease the value
• BACK : To go back to previous function or escape the
current function
• Lid sensor-
Instrument has four mode -
• Start New Experiment
• Continue Previous Experiment
• View logs
• PC connectivity
1. Start New Experiment: To start with new exp.
- Press OK once Start
- New Exp. appears on the screen
• Set cut off time:-
• Set temperature :
• Heating aluminium plate :
• The Screen will show heating, please wait to indicate
heating is going on
• Exp. indicating the experiment number
• S. No. indicating the Sequence Number
• S. Temp indicating Set temperature value
• P. Temp indicating present plate temperature value
• On opening the lid of the chamber, the green light
marked as LID on the control panel will glow
indicating that lid is open
• Indication to remind the user to close the lid to
avoid temperature fluctuations
• Temp - light will keep blinking till the set
temperature is achieved
• Once the plate temperature is equal to set
temperature value, the display will show place
animal
• Place the animal on the plate surface and press foot
switch
• Keep foot switch pressed till the reaction time is
noted
• Observe the reaction time by noting the jumping
response or the licking of paws
• As soon as you observe the jumping response, release
the pedal switch so as to stop the timer and note
down the reaction time
• Once the foot switch is released after watching
reaction time, reading for reaction time will be
saved in the log
• Test compounds are administered i.p. and latency
period is recorded after 20, 60 and 90 min

data transfer
Tramadol in hot plate analgesiometer
• Dose – 20 mg/kg
• Route - intraperitoneal
Procedure : during testing
• Weigh the animals and mark properly to distinguish
from one another
• Divide animals into two groups (n = 6 in each group)
Group 1:
Control group
Rats are given saline at the equivalent dose of drug
Group 2:
Treatment group
Rats are given Tramadol (20 mg/kg)
Weigh the animal and mark properly to distinguish from
one another
Put the animals on hot plate analgesiometer
Observe the animals for response (Paw licking or
jumping)
Make the observation table

Data analysis and results
• The following response are observed to note the
reaction time:
- Paw licking
- Jumping response
Precaution:
• Always note that the lid of plate is enclosed to get accurate and
fast temperature
• If plate temperature fluctuates, wait till the plate temperature
is achieved to set value
• Clean the paw and hot plate for uniform temperature
distribution
