After doing the plates last week, we identified the tubes and added colonies in the tubes, according to the protocol said. Two colonies were heated and two were left at room temperature. We prepared gel electrophoresis to examine the different pieces that were to be observed with UV light to determine if the GFP was located in the gel. When it came time to add the gel solutions, we had many problems. The solution was very heavy and did not enter the tip of the pipette. The same thing happened when we used larger pipette tips. After completing the protocol, only group # 3 completed the experiment, and we could observe the GFP in lane 9 of the gel. <br />
