Studies of antibacterial effect of STS Conditioner with corn mash samples from corn ethanol plants. • The goal was to find the lowest concentration of STS CONDITIONER that will kill bacteria but not impede the growth or fermentation efficiency of the yeast. • We performed studies with the corn mash sample from Prairie Horizon. – The corn mash sample was taken prior to the prop so we needed to add yeast. – Before adding anything we tested for growth and found two populations that seem to be bacteria and a fungus. – The latter has a phenotype that is distinctly different from bacteria and S. cerevisiae. • We determined values for MIC of STS CONDITIONER for the bacteria and the other contaminant in this sample to be 0.05 and 0.5, respectively. • We added yeast and STS nutrients and determined the MIC of STS CONDITIONER for the yeast. • The results show that STS CONDITIONER impacts yeast propagation at levels of 0.5% and above, even in the presence of STS nutrients.
Studies of antibacterial effect of STS Conditioner with corn mash samples from corn ethanol plants. • Following this series we spiked a sample with 2 x 106 cells of bacteria (lactobacillus fermentum) and ran yeast (Saccharomyces cerevicea) propagation trials in the presence of various concentrations of STS Conditioner, Lactrol, and Isostab. • The concentrations of Lactrol and Isostab were chosen based on the recommended values (Lactrol 0.5 ppm, Isostab 50 ppm) in the product literature. • Controls involving yeast only and bacteria only were also included. The concentrations of the test solutions were: STS 0.5 % 0.1% 0.05% Conditioner (5,000 ppm) (1,000 ppm) (500 ppm) Lactrol 1 ppm 0.5 ppm 0.25 ppm Isostab 100 ppm 50 ppm 25 ppm
Studies of antibacterial effect of STS Conditioner with corn mash samples from corn ethanol plants. • Total number of baffled flasks - 12 • Final volume in flasks: 50ml = 40ml (w/v) corn mash + 10ml dewater • pH of the corn mash + yeast samples was measured before and after addition of the test solutions, and also at 3 subsequent time points for all samples • Ave initial pH 5.9 for all samples before the addition of test solutions • pH for Lactrol and IsoStab decreased on average to pH 5.3 • pH for STS Conditioner @ 0.50% increased 4.58 to 5.3 @ 0.10% increased 5.23 to 5.3 @ 0.05% decreased 5.78 to 5.35
Studies of antibacterial effect of STS Conditioner with corn mash samples from corn ethanol plants. • For the case of Lactrol addition to samples containing yeast and bacteria, photos of the plates at the three Lactrol concentrations were shown. • Figure 1, show a slight decrease in the colonies (presumably a decrease in bacterial colonies) as a function of Lactrol concentration. – Little or no variation was observed for Isostab in a similar trial. • For the case of STS Conditioner addition to samples containing yeast and bacteria, photos of the plates at the three STS Conditioner concentrations are shown in Fig 2.Figure 1 Figure 2
Studies of antibacterial effect of STS Conditioner with corn mash samples from corn ethanol plants. • Qualitatively, from the growth on the plates it is visually apparent that STS Conditioner at 0.5% is more effective at killing the bacteria than Lactrol at 1 ppm under these conditions. – However, when both the yeast and lactobacillus are present at high numbers so it is difficult to distinguish them and make quantitatively accurate counts.
