Polyclonal Antibody Development Service MTR offering a variety of Polyclonal Antibody Services to fit your research and production needs. we develop polyclonal antibodies to targets including parasites, cancer-specific surface antigens, hormones, growth factors and their receptors, glycoproteins, peptides, proteins and their modification sites (mutation site-specific, phosphorylation site-specific, cleavage site-specific, etc), and other antigens such as small-molecule compounds and DNA. We have created antibodies for various applications, such as functional assays, antibodies for immunoprecipitation, IHC, IF, flow cytometry, western blot, and EIA. Project Timeline: 4-6 weeks Deliverables: We will deliver approximately 1 ml of mice serum or 5 ml of Rabbit serum with guaranteed titer at least 1:10,000 (Range 1:10,000 to 1:100,000). 2. Monoclonal Antibody Development Services MTR offer mouse hybridoma development technology for producing custom monoclonal antibodies from synthesized peptide or recombinant protein antigens. Novel immunization strategies, antigen design, and screening technologies are essential for the successful generation of monoclonal hybridoma cells lines. Our immunization protocols take advantage of using altered immunization routes, combinations of ligand carriers and dosing strategies to produce the most diverse monoclonal antibody population possible. Antigens must be carefully designed, especially when dealing with peptides, to generate a robust reagent that is able to recognize conformational changes of the native protein in assay conditions. Antibody and Protein Purification Services Services and Offerings MTR provide purification service antibodies and protein utilizing a wide array of protein purification methods; from research (mg) to scale-up (grams) quantities. We also offer low endotoxin production for in-vivo use. Our goal is to find or develop the antibody purification procedure which provides the best balance between yield, purity, and cost. Available Antibody Purification services and methods include: Ammonium Sulphate Precipitation This method is useful for concentration and partial purification of antibodies from most sources and all species. Although, on its own the yields of antibody are impure, it is the method of choice when combined with ion exchange for large volumes of anti-sera. It is not recommended for purification of tissue culture supernatant. Protein A This process binds the Fc region of major sub-classes of mouse and rat IgG to varying degrees. The results with protein A are usually high purity and high yields. It is useful for large-scale purifications of tissue culture supernatants. Protein G Protein G can be used for purifying antibody from most sources. Like protein A, the results are usually of high purity and high yield. It is applicable to a wider range of immunoglobulins than protein A, but elution procedures can be much harsher.
  3. 3. Ion Exchange Chromatography This method is used for all sources of antibodies. It is applicable to all lgG isotypes. It is usually the method of choice when purifying antibodies from ascites, as the monoclonal can be resolved from the host immunoglobulins. Antigen Affinity This is the method of choice when specific antibody is required from polyclonal anti-sera. It requires pure antigen for the best results and can sometimes result in inactive antibody. Size Exclusion This method is appropriate for IgM antibodies from all sources. It is used when IgM Antibodies need to be separated from polyclonal anti-sera. It is not recommended as a single stage. Project Timeline: 2-3 weeks Deliverables: Purified antibody Project report 4.
