      • Characteristics: Gram Negative, rod-shaped gliding bacteria. Occurs widely in soil and water.
      • Research Focus: Identification of components of the gliding machinery of F. johnsoniae
      • Research Methodology: A genetic approach to characterize all the genes and proteins involved.

      December 2006 – August 2008 Characterization of the gldN and gldO genes in gliding motility of Flavobacterium johnsoniae. Created the first double knockout mutant in the gld genes, which play a vital role in gliding motility. The gldNO knockout mutant was generated using a novel phage selection method. Complementation and phenotypic analysis confirmed their involvement indicating that GldN and GldO are partially redundant components of the motility machinery.

      May 2005 - December 2006. Characterization of spr E gene of Flavobacterium johnsoniae in gliding motility & chitin utilization. Transposon mutagenesis was used to isolate the sprE mutants. The sprE and the downstream fjoh_1052 genes were successfully cloned into DNA constructs . Complementation studies demonstrated the involvement of sprE in gliding motility. Bioinformatic analysis predicted SprE to be a lipoprotein with six TPR domains, which are likely to be involved in protein-protein interactions.

      January 2005 - May 2005. Complementation analysis of spontaneous point mutants of Flavobacterium johnsoniae using gld A gene. A bank of 37 different point mutants was screened for mutation in gld A gene and checked for restoration of the wild type phenotype. One of them was successfully complemented indicating the presence of a gld A point mutation.
