Characteristics include: gene expression patterns associated with the down-regulation of genes involved in cell death and survival; high expression of homing/migration chemokines.

We recovered humanCB-MSCs fromcryopreservationandbriefly culturedthesecells (48hours)at20% O2-5% CO2inMSC-Gro,low-serum medium(Neuromics, SC00B1).Asa stressfulenvironment,thesecells were thentransferred to1% O2-5%CO2at 105/ml innutrient-poormedium (expiredRPMI)supplementedwith 0.12PZ/mlcollagenaseNB4(Serva, catalognumber17454) at4oC. We retrieved viablecells 48-hourslaterand transferredthemtoMSC-Gro humanized,completemedium (Neuromics’,SC00B2).Theimages aboveshowformationofmulti-cell clusters~50 to130 umat2days(Left) and4days(Rt)followingexposureto stress.   5. 5. Images: (A) Clusters of stress- selected cells are positive for SSEA-3, a pluripotency marker. Following blockage of non- specific binding, cultures were incubated with a Alexa Fluor- conjugated SSEA-3 monoclonal antibody (MC-631-AF 488; BioVision, catalog number 3068-25T) at 10 ml/ml overnight. (B)SSEA-3 positive cells within cell clusters together with propidium iodide. Red cells are non- viable. SSEA-3 is specific to embryonic stem cells; MSCs were negative for SSEA-3.
