  • 1. Live Webinar: Elisa Data Analysis with MasterPlex ReaderFit Robert Lynde and Allen Liu September 10 th , 2009
  • 2. Overview 1) File Import Wizard 2) Automatic Detection of Outliers 3) Marking Wells and Grouping of Replicates 4) Entering Standard Information 5) Best Fit and Best Fit Parameters 6) Plotting Unknown Data Points on the Standard Curve 7) WYSIWYG Data Table 8) Chart Customization 9) Templates 10) Custom Reports 11) Virtual Plate
  • 3. File Import Wizard
    • The File Import Wizard can recognize and import text & Excel files with raw data 2 different formats:
      • Plate
      • List
    10/22/09
  • 4. Automatically Detect Outliers
    • Mark and select thresholds based on:
      • Raw signal (e.g. absorbance, luminescence, fluorescence, RLU etc.)
      • Concentration
      • Standard Deviation
    • Options are located under Plate Preferences->Plate tab
  • 5. Marking Wells & Grouping of Replicates
    • There are 4 different well types:
      • Background
      • Standard
        • Auto Fill Feature – works really nicely for serially diluted standards
        • Manual Input – one can also type in the values
      • Unknown
      • Control
    • The following stats will be calculated based on your replicates (for both raw signal and Concentration):
      • Standard Deviation
      • % CV
      • Mean
    • Bonus Tips
      • Group replicates with the group stamping feature
      • Pasting sample names
      • Batch dilution factor for unknowns
  • 6. Best Fit and Best Fit Parameters
    • Leave it to the software to do all the hard work for you with the Best Fit feature
    • Optimize based on your chosen parameter with the default being RMSE
    • Optimize the Best Fit based on:
      • Root mean square error (RMSE)
      • R-square
      • Standard deviation of % Recovery
    • Options are located under Plate Preferences->Plate tab
  • 7. Plotting Unknown Data Points on the Standard Curve
    • Get a quick visual overview of where your unknown data points fall on the standard curves
    • Useful for determining whether your standard range is sufficient
    • Useful for determining whether the unknown data points fall onto to the linear or dynamic range
    • Bonus Tip – Zoom in and out on standard curve chart using the Shift and Alt keys
  • 8. WYSIWYG Data Table
    • Customize the Data Table by:
      • Adding/Removing data columns
      • Filtering data
      • Sorting data
    • And then simply export to many different output formats including Excel
  • 9. Chart Customization
    • Easily change and configure many chart properties such as:
      • Legends
      • Axes
      • Labels
      • General Appearance
    • Bonus Tip – Sample order on X axis
  • 10. Templates
    • Save all your work into templates and reload them in seconds
    • Templates are available for:
      • Plate layout and standard information
      • Data Table
      • Charts
  • 11. Custom Reports
    • The Data Table is extremely flexible but it is not the one-size-fits-all solution
    • This is where the Custom Report feature leaves off
    • One free custom report with purchase and additional reports available as a service
  • 12. Virtual Plate
    • Easily merge different plate files together with the Virtual Plate feature
    • Handy for:
      • Using standards on one plate for another plate
      • Stitching together 2 incomplete files from the same run
      • When 96 wells just is not enough for your assay
  • 13. Resources
