4 Parameter Logistic (4PL) A = minimum asymptote D = maximum asymptote C = inflection point B = slope factor/steepness of curve < low standard OR lowest extrapolated value > high standard OR highest extrapolated value

Caution on Extrapolation tiny change in MFI = HUGE change in concentration

Assumptions of the 4PL & 5PL
2 assumptions about how the data are scattered around the curve…
The scatter follows a Gaussian or “normal” distribution
The standard deviation of the scatter (of the response values or MFI) is the same for all values of x (or concentrations in our case)

Heteroscedasticity

Weighting
1/Y^2 (Relative weighting) – Most commonly used
1/Y – Most effective when the scatter follows a Poisson distribution
Mainly affects points on the lower portion of the curve
Test by doing before/after on the % Recovery
