Transcript of "Flexigas Symposium 2013 - Parameters of biogas production: the microbial connection - Gert Hofstede"

  1. 1. Parameters of biogas production:the microbial connectionGert J.H.Hofstede, Expertise Centre ALIFE, Institute for Life Sciences and Technology,HanzeUniversity of Applied Sciences, Groningen, the Netherlands1
  2. 2. Parameters of biogas production:the microbial connection2
  3. 3. Parameters of biogas production: the microbial connection33Dr. Ir. Jan Peter Nap(lector)Dr. Folkert Faber(senior lecturer)Brian Wouterse(trainee)
  4. 4. Parameters of biogas production:the microbial connectionIn the Netherlands: About ~180 biogas installations. In these installations manure and residualmaterials are (co-)digested. 0,5 billion (=109) m3 biogas is produced Green electricity can be generated.Current situation:4
  5. 5. Parameters of biogas production:the microbial connectionCurrent situation: Relatively limited knowledge on the complexcommunity of microorganisms The economy of biogas plants does not allowfailure (cleaning and restarting) Fairly conservative enterprise5
  6. 6. Parameters of biogas production:the microbial connectionWhy this research ? The use of defined digestate of microorganismstargeted towards the feedstock might speed upbiogas production. Opening the microbial black box is supposed toincrease biogas yield and the overall processefficiency. The overall objective of this research is to describethe microbial community that participates in the AD6
  7. 7. Parameters of biogas production:the microbial connection Have different biomasses distinct biomethanepotentials? Are different methanogenic communities, responsible for biogas production? Is the methanogenic community influenced bydifferent environments and different processparameters?Research question7
  8. 8. Parameters of biogas production: the microbial connectionExperimental tools Automatic Methane Potential Test System(AMPTSII)  batch 2 Labscale bioreactor (10 l, Infors) continuous Molecular techniques (qPCR )8
  9. 9. Parameters of biogas production: themicrobial connectionAMPTSII, bioprocescontrol9
  10. 10. Parameters of biogas production: the microbial connectionAMPTSII-experiments: Reproducibility 3 different digestates 2 types of biomass10
  11. 11. Parameters of biogas production: the microbial connection General setup: 15 bottles, 1 type of biomass(maize), 1 type of digestate (Proces B.V) ,30 days Digestate to biomass ratio of 1:1 (based on% Organic Dry Matter [ODM]) was used.General set up ofAMPTSII assays11
  12. 12. Parameters of biogas production: the microbial connectionConclusion The AMPTSII yields reproducibleresults, both in terms of total gasvolume and in terms of methaneproduction12
  13. 13. Parameters of biogas production: the microbial connectionConclusion: No significant difference betweentotal biogas productions frommaize by different types of digestates( Donderen, Lutjegast en Tolbert). (ANOVA, one-tail, p=0.05).13
  14. 14. Parameters of biogas production: the microbial connectionFuture-plans: In future experiments, we will look thebiogas flow-rate with for example‘biomass adapted digestates’  thedigestate (Proces B.V) was notadapted to maize.14
  15. 15. Parameters of biogas production: the microbial connectionFuture-plans:15
  16. 16. Parameters of biogas production: the microbial connectionBiogasplants (F1 and F2)16
  17. 17. Parameters of biogas production: the microbial connectionThe overall objective of this research is todetermine the relative changes in microbialcommunity that participates in the anaerobicdigestion on a Labscale Infors bioreactor17
  18. 18. Parameters of biogas production: themicrobial connectionMethanogens, the biological generators of methane18
  19. 19. Parameters of biogas production: the microbial connectionFastRNA® SPIN KIT for SOILTotal DNA and RNA isolation19
  20. 20. Parameters of biogas production: the microbial connectionPrimer combinations used in this studyfor monitoring the populationdynamics of methanogens in biogasplants20
  21. 21. Parameters of biogas production: the microbial connection This study revealed the limitation of the qPCRdue to the primers.  in silico by the RDP-site Experimental circumstances probablyinfluence the ‘reference’ Archaea populationConclusion/discussion Commonly used primers in literature shouldbe reconsidered in the light of expanding datain RDP21
  22. 22. Parameters of biogas production: the microbial connection Since available primers may not be applicableto all samples, there is a requirement to designmore primers to ensure adequate coverageand amplification of methanogens.22Conclusion/discussion
  23. 23. Parameters of biogas production: the microbial connection Therefore, we also look to at anotherapproach: mRNA of (key-) enzymes that play arole in the methanogenesis :23 Together with Marina Gil Lopez from theUniversitat de Barcelona we did the firstexperiments on RNA
  24. 24. Parameters of biogas production: the microbial connection Identification of Enzymes Involved in synthesisof biogas The challenge is to find (key-)enzymes thatcan be linked to (specific methanogenic)micro-organisms. as key enzymes can be linked to micro-organisms, relative amounts of micro organismscan be calculated.24
  25. 25. Parameters of biogas production: the microbial connectionThe future for me? I will end up with abucket full of digestate (or micro-organisms) optimized for every biomass. That is applied science in my opinion25
